| It is important and useful for highly effective, economical and safe use of herbicides to screen and breed of herbicide-tolerant rice and find effective additives. Our recent results indicated that japonica rice show higher sensitivity to bispyribac-sodium compared to indica rice. Futher analysis found that there are differences between indica and japonica rice, such as the activity of glutathione S-transferase (GST), contents of cytochrome P450 and cytochrome b5, relative gene expression of GSTU9 and CYP72A23 after treated with bispyribac-sodium. Thus, the different amounts of GSH, GSSG or CytP450 between indica and japonica rice were determined after treated with bispyribac-sodium. the differences of GST and CytP450 were measured between indica and japonica rice, such as the activities of GST and CytP450,the gene sequence, gene expression and protein level of GSTU9 and CYP72A23, in order to elucidate the role of GST and CytP450 in different sensitivities of indica and japonica rice. Combining with related mutants, the function of GSTU9 and CYP72A23 in different sensitivities of indica and japonica rice would be further investigated in this project.The activity inducers or inhibitors were screened for effective use of bispyribac-sodium The results would be contributed to explore the metabolism and increase the application of bispyribac-sodium. |

培育篩選耐除草劑的水稻材料以及發(fā)現(xiàn)有效助劑,是除草劑高效、經(jīng)濟(jì)和安全使用的重要保證。我們前期工作發(fā)現(xiàn),秈稻和粳稻對除草劑雙草醚的敏感性不一;進(jìn)一步分析發(fā)現(xiàn),秈稻和粳稻的谷胱甘肽硫轉(zhuǎn)移酶(GST)活性和細(xì)胞色素P450氧化酶系(CytP450)含量不一。本項(xiàng)目擬以粳稻日本腈和秈稻9311為試材,比較雙草醚脅迫下GST反應(yīng)底物(GSH、GSSG)和CytP450含量差異時(shí)序變化;通過體內(nèi)和體外測定,比較應(yīng)答對雙草醚的酶活特性;比較GSTU9和CYP72A23基因和其編碼產(chǎn)物的結(jié)構(gòu)差異和應(yīng)答雙草醚的表達(dá)特征;通過遺傳突變體,揭示GSTU9和CYP72A23在耐受雙草醚中的貢獻(xiàn);添加酶活誘導(dǎo)劑或抑制劑,分析雙草醚進(jìn)一步高效應(yīng)用的有效助劑。以上研究結(jié)果,不僅對于揭示秈稻與粳稻中GST和CytP450對雙草醚敏感性差異的機(jī)理具有科學(xué)意義,而且為水稻田中高效安全應(yīng)用雙草醚提供科學(xué)依據(jù)。 |
