【研究方向】development of smart lanthanide-luminescence probes for biomedical raman analyses and imaging
【課題簡介】
the project aims to develop a new methodology of cell imaging, by combining lanthanide-luminescence and raman microscopy. this approach overcomes a notorious problem encountered in raman imaging of small molecules whose spectroscopic signals are masked by those of other biomolecules. the probes’ emission bands will fit into the cellular‘silent’ range of the raman spectrum, thus greatly increasing ‘chemical’ contrast in the compound-specific (raman) images of living cells. compared to currently available (non-luminescent) raman tags our‘prototype’probe offers much stronger luminance, by ca two orders of magnitude. this is of course instrumental in analyzing tiny cellular structures. we will tag the probes to specific biomolecules, in order to target them to specific sites in living cells.
an important objective of the project is to design and develop efficient lanthanide probes suitable for chiral sensing of biomolecules such as proteins and nucleic acids. novel lanthanide complexes with a specific binding group will be synthesized and characterized, and their chiral spectroscopic response upon interaction with proteins and nucleic acids monitored by roa-cpl methodology.
