|
桑科中藥類酪氨酸酶抑制劑研究 中藥酪氨酸酶抑制劑的研究和開發(fā)是當(dāng)今研究熱點。酪氨酸酶與人的衰老、傷口愈合、昆蟲的發(fā)育及果蔬的褐變有密切關(guān)系。故對于酪氨酸酶的研究具有廣泛的理論意義和實際應(yīng)用價值。如美白化妝品的研究方面,酪氨酸酶是皮膚中黑色素合成的起始酶,如果蔬保鮮劑的研究方面,酪氨酸酶會影響果實的褐變過程,選取中藥天然成分作為酪氨酸酶抑制劑滿足人們對于“綠色化妝品、回歸大自然”產(chǎn)品的要求。 本課題選擇?浦兴幧0灼ぁ⑸H~、桑枝、桑葚作為研究體系,考察其抑制酪氨酸酶作用,進(jìn)而為酪氨酸酶抑制劑的開發(fā)和應(yīng)用提供依據(jù)。 研究中,分別以L-酪氨酸和L-多巴作為底物,首先考察了四味桑科中藥抑制酪氨酸酶單酚氧化酶和二酚氧化酶的能力。實驗結(jié)果顯示,桑科中藥對于單酚氧化酶的抑制比較為:桑枝﹥桑白皮﹥桑葉﹥桑葚。不論在低濃度還是在高濃度下?浦兴幰种贫友趸傅谋容^為:桑白皮﹥桑葉﹥桑枝﹥桑葚;桑白皮水提取物的IC50值為1.13 mg/mL,桑葉水提取物的IC50值為2.14 mg/mL,桑枝水提取物的IC50值為6.64 mg/mL,桑椹水提取物的IC50值為12.73 mg/mL。?浦兴幥宄蹶庪x子自由基能力比較為:桑葉﹥桑白皮﹥桑葚﹥桑枝,其IC50值分別為:5.18 mg/mL、5.91 mg/mL、6.58 mg/mL和11.01 mg/mL。綜合評價得出桑白皮為桑科中藥最優(yōu)酪氨酸酶抑制劑。最后,用超氧陰離子自由基、DPPH自由基和ABTS自由基評價了四味?浦兴幟腊啄芰?寡趸蛩乜疾爝^程中發(fā)現(xiàn)桑葉的抗氧化作用比較穩(wěn)定且高效,桑白皮次之;桑枝的抗氧化作用最差;桑椹的抗氧化效果具有選擇性,對清除DPPH自由基尤為突出。通過Lineweaver-Burk雙倒數(shù)作圖法確定了桑白皮水提物抑制酪氨酸酶的機制類型為混合型抑制劑,其抑制常數(shù)(KI,KIS)分別為4.34 mg/mL和23.26 mg/mL。其次,桑白皮對草莓保鮮作用研究結(jié)果顯示,保鮮液能有效地降低草莓的爛果率、失重率并減緩草莓中維生素C和總糖含量的流失,但受溫度影響較為嚴(yán)重,對于草莓中PPO、POD酶的影響也為復(fù)雜。綜上研究發(fā)現(xiàn),桑白皮為?浦辛己玫睦野彼崦敢种苿,在果蔬保鮮和皮膚美白等方面有廣闊的應(yīng)用價值。 關(guān)鍵詞:桑科中藥、酪氨酸酶、抗氧化、抑制機制、果蔬保鮮劑 翻譯器翻譯的勿擾,謝謝! |

Sankoh tyrosinase inhibitor class of Chinese medicine Tyrosinase inhibitor medicine research and development in today's hot topic. Such as whitening cosmetics research, tyrosinase is the synthesis of melanin in the skin starting enzyme, if the vegetable preservative research, the tyrosinase activity will affect the browning of fruit, selected natural ingredients of Chinese medicine as a tyrosinase inhibitor meet people for "green cosmetics, return to nature" product requirements. The topics chosen for Sankoh Chinese Morus alba, mulberry, mulberry, mulberry as a research system to study its role in inhibition of tyrosinase, tyrosinase inhibitors and thus to the development and application of basis. Study, respectively, L-tyrosine and L-DOPA as a substrate, the first taste of Sankoh investigated four Chinese inhibit tyrosinase and two single-phenol oxidase and polyphenol oxidase capacity.
