我的導(dǎo)師看過之后說英語(yǔ)有些問題,但是本人才疏學(xué)淺,實(shí)在不會(huì)了,求高手幫忙調(diào)整一下順序,或者那個(gè)詞、詞組用的不對(duì)幫忙更換一下,不勝感激!本人新手注冊(cè)時(shí)間尚短,奉獻(xiàn)全額金幣,望答復(fù)!~ 摘 要 本文通過活性篩選,選用人肝癌Hep G2 細(xì)胞株用他克林誘導(dǎo)細(xì)胞毒性,將烏蘇里瓦韋(Lepisorus ussuriensis)各溶劑浸膏作用于經(jīng)他克林處理過的Hep G2細(xì)胞,結(jié)果顯示烏蘇里瓦韋乙酸乙酯浸膏能夠有效的降低他克林對(duì)Hep G2細(xì)胞產(chǎn)生的毒性,具有較好的細(xì)胞保護(hù)作用。 烏蘇里瓦韋乙酸乙酯浸膏能夠減少脂多糖(LPS)/D-氨基半乳糖(D-GalN)引起的急性肝損傷小鼠的死亡率。能顯著降低由LPS/D-GalN引起的肝損傷小鼠的谷丙轉(zhuǎn)氨酶(ALT)和谷草轉(zhuǎn)氨酶(AST)活性,降低丙二醛(MDA)含量,升高超氧化物歧化酶(SOD)活性。結(jié)果顯示烏蘇里瓦韋乙酸乙酯浸膏通過抗氧化活性對(duì)LPS/D-GalN所致的急性肝損傷小鼠具有保護(hù)作用。 綜合應(yīng)用多種層析分離技術(shù),從烏蘇里瓦韋中分離得到6個(gè)化合物,并進(jìn)一步通過波譜分析(1H-NMR,13C-NMR等)鑒定了6個(gè)化合物的結(jié)構(gòu)分別是:2-丙烯酸-3-(4-羥基苯基)-乙酯(1),4-乙氧基苯甲酸(2),圣草酚-7-O-葡萄糖苷(3),4’,5,7-三羥基-3-甲氧基黃酮-7-O-β-D-葡萄糖苷(4),3’,4,5,7-四羥基-3-甲氧基黃酮-7-O-β-D-葡萄糖苷(5),2-(3,4-二羥基苯基)乙基-6-[3-(3,4-二羥基苯基)-2 -丙烯酸]- β-D-葡萄糖苷(6)。 其中化合物1-6均能有效的降低他克林對(duì)Hep G2細(xì)胞產(chǎn)生的毒性,具有較好的細(xì)胞保護(hù)作用。 目前對(duì)烏蘇里瓦韋肝保護(hù)活性成分的研究仍然較少。因此,分離烏蘇里瓦韋肝保護(hù)活性成分,并進(jìn)一步確定其肝保護(hù)活性成分的結(jié)構(gòu)有著重要的理論和實(shí)際意義。 關(guān)鍵詞:瓦韋;動(dòng)物實(shí)驗(yàn);結(jié)構(gòu)鑒定;細(xì)胞實(shí)驗(yàn) Abstract In vitro screen assay, use of human hepatoma Hep G2 cell line induced by tacrine cytotoxicity, the L.ussuriensis role in the solvent extracts of tacrine treated by Hep G2 cells, The results showed that ethyl acetate extract of L.ussuriensis can effectively reduce tacrine on the toxicity of Hep G2 cells. Ethyl acetate extract of Lepisorus ussuriensis bring down the activity of AST and ALT in the LPS and D-GalN induced hepatic serum, bring up the liver SOD activity, reduced the content of MDA, and improve the survival rate of mice, which has significan protection to hepatic mice. Experimental study on L.ussuriensis the the chemical composition, using various separation methods, 6 compounds were isolated from the L.ussuriensis and further analyzed by spectral Identified the structure of 6 compounds were : 2-propenoic acid-3-(4-hydroxyphenyl)-ethyl ester(1), 4-ethoxybenzoic acid(2), eriodictyol-7-O-glucoside(3), 4’,5,7-trihydroxy-3-methoxy flavone-7-O-β-D- glucose -ide(4), 3’,4’,5,7-tetrahydroxy -3-methoxy flavone-7-O-β-D- glucoside(5), 2-(3,4-dihy-droxyphenyl)ethyl-6-[3-(3,4-dihydroxyphenyl)-2-propenoate]-β-D-glucoside(6). Compounds 1-6 isolated from the L.ussuriensis can effectively reduce toxicity of Hep G2 cells. Current research on Liver protection active ingredient of L.ussuriensis is still not popular. Therefore, it is of important theoretical and practical significance on the separation of Liver protection active ingredient of L.ussuriensis and further identify the structure of Liver protection active ingredient. Abstract In vitro bioactivity assay, cytotoxicity was induced by tacrine with human hepatoma cell line Hep G2. Extracts of Lepisorus ussuriensis with various organic solvents were applicated in treatment of Hep G2, which was treated as above-mentioned. Results indicated that ethyl acetate extract effectively reduced cytotoxicity of Hep G2 caused by tacrine and thus played a role in cell protection. Ethyl acetate extract of Lepisorus ussuriensis lowers death rate of acute hepatic injury mice caused by decrease of LPS/ D-GalN, lowers significantly activities of ALT and AST of hepatic injury mice caused by LPS/D-GalN and reduces content of MDA, as well as bring up activities of SOD. Results indicate through activities of antioxidant ethyl acetate extract of Lepisorus ussuriensis play a role in cell protection of acute liver injury mice caused by LPS/D-GalN. Various isolation techniques were applicated. Six compounds were isolated from Lepisorus ussuriensis, which were characterized through 1H-NMR to be 2-propenoic acid-3-(4-hydroxyphenyl)-ethyl ester(1), 4-ethoxybenzoic acid(2), eriodictyol-7-O-glucoside(3), 4’,5,7-trihydroxy-3-methoxy flavone-7-O-β-D- glucose-ide(4), 3’,4’,5,7-tetrahydroxy -3-methoxy flavone-7-O-β-D- glucoside(5), 2-(3,4-dihy-droxyphenyl)ethyl-6-[3-(3,4-dihydroxyphenyl)-2-propenoate]-β-D-glucoside(6). Among them, all compounds can effectively reduce cytotoxicity of Hep G2 caused by tacrine and thus play a role in protection of cells. Currently, relatively less is reported on Liver protection active ingredients. Therefore, it is of theoretical and practical value to further isolate and characterize liver protection active ingredients of L.ussuriensis.
