分別提取豬18種組織的RNA，反轉(zhuǎn)錄為cDNA，利用RT-PCR方法擴(kuò)增腫瘤壞死因子（TNF-α）基因編碼區(qū)序列，將純化的片段與pMD18-T載體連接，轉(zhuǎn)化大腸桿菌DH 5α，篩選陽性克隆并測定其序列，利用生物信息學(xué)方法分析其序列特征，并與其他物種構(gòu)建系統(tǒng)進(jìn)化樹。同時采用半定量RT-PCR方法分析TNF-α基因在豬18種組織中的表達(dá)情況。結(jié)果顯示，豬TNF-α基因編碼區(qū)全長699 bp（GenBank登錄號：JF831365），其編碼232個氨基酸。氨基酸序列同源性分析表明，與貓等12個物種具有80%以上的相似性，分子系統(tǒng)進(jìn)化研究表明與牛、山羊、綿羊及海豚的關(guān)系較近。多組織半定量RT-PCR研究表明TNF-α mRNA在豬的多個組織中均有表達(dá)，其中在中腦、卵巢、脾、肺、神經(jīng)及胃中表達(dá)量較高。 非誠勿擾�。。�！ |

We extracted RNA from 18 kinds of pig tissues separately，anti-transcripted them to cDNA，and used RT-PCR method to amplify the gene code aera sequences of tumor necrosis factor，then jointed the purified sequences with pMD18-T carrier to transform E.coli. Positive clones were screened and their sequences were determined.Using bio-information method to analyse their characters，system evolution tree was set up with other species.Meanwhile,we used semi-quantitative RT-PCR method to analyse the expression of TNF-α gene in 18 kinds of pig tissues . The result showed the whole length of pig TNF-α gene code aera was 699 bp（GenBank accession number：JF831365 ），it coded 232 amino acids. The amino acid homology analysis showed it had over 80% homology with 12 species like cat，molecular system evolution research indicated it had a close relation with cattle，goat，sheep and dolphin.Multi-tissues semiquantitative RT-PCR study showed TNF-α mRNA expressed in several tissues of pig，and its expression was higher in brain，ovary，spleen，lung，neuro and stomach. 新手，不足之處還望大家多多指教 |
