AJHG：基因突變致少數(shù)人天生沒指紋
|

20世紀(jì)30年代臭名昭著的劫匪約翰-狄林杰為了不讓警方發(fā)現(xiàn)犯罪現(xiàn)場(chǎng)的指紋與他的指紋一樣，不惜忍受巨大的痛苦，利用硫酸把指紋燒掉。但是對(duì)我們來說，沒有指紋是一件非�？膳碌氖虑椋鼤�(huì)在邊界控制和證明身份時(shí)造成很多麻煩。科學(xué)家目前已經(jīng)確定可以導(dǎo)致一些人天生沒有指紋的基因。 皮紋病患者天生沒有指紋 這種情況被稱作皮紋病(adermatoglyphia)或者“入境延期病(Immigration Delay Disease)”，患有這種疾病的人天生手指肚上沒有指紋。以色列特拉維夫大學(xué)的艾里-斯普雷徹教授獲得的最新發(fā)現(xiàn)顯示，一種基因突變?cè)斐蛇@種與眾不同的病變。一名瑞士女性試圖穿越邊境，進(jìn)入美國(guó)，邊境控制人員需要收集她的指紋。然而當(dāng)這位女子告訴他們，她無法滿足他們的要求，因?yàn)樗緵]有指紋時(shí)，這些官員感到非常迷惑。這也促使醫(yī)學(xué)界開始注意皮紋病。 研究人員對(duì)這位女性以及她的另外9名沒有指紋的家庭成員進(jìn)行遺傳分析。特拉維夫大學(xué)的科學(xué)家將存在這種情況的人的基因與沒有這種情況的人的基因進(jìn)行比對(duì)，確定基因變異到底發(fā)生在哪里。他們發(fā)現(xiàn)，SMARCAD1突變影響了指紋的形成。研究發(fā)現(xiàn)，沒有指紋的人擁有的與皮膚發(fā)育有關(guān)的這種基因更少�，F(xiàn)在科學(xué)家將能進(jìn)一步研究這種基因是如何調(diào)控指紋的發(fā)展的。 與DNA一樣，每個(gè)人的指紋也是獨(dú)一無二的，即使是同卵雙胞胎也不例外，因此它們也成為破案和國(guó)際旅行管理等的重要鑒定工具。它們之所以會(huì)被當(dāng)作確定身份的工具，是因?yàn)樗鼈冊(cè)谑芫?4周后就會(huì)發(fā)育健全，而且整個(gè)一生都不會(huì)發(fā)生任何改變。目前全球僅有4個(gè)記錄在案的家庭存在這種情況。斯普雷徹表示，不僅手指上長(zhǎng)有帶圖案的皮膚，手掌、腳趾和足底也長(zhǎng)著被稱作皮紋的紋路。然而他說：“胚胎發(fā)育階段導(dǎo)致指紋形成和擁有不同圖案的因素目前大部分還不得而知�！� 除了缺少指紋外，這種情況還導(dǎo)致汗腺減少。指紋異�？赡芤彩浅霈F(xiàn)更嚴(yán)重的疾病的一種預(yù)警信號(hào)。該研究成果發(fā)表在《美國(guó)人類基因?qū)W期刊》上。2009年，一名中國(guó)女性通過整形手術(shù)改變她的指紋，想利用這種方法非法進(jìn)入日本。東京警方表示，以前因簽證過期被驅(qū)逐出日本的林榮(Lin Rong)花了1.5萬美元在中國(guó)做了這項(xiàng)整形手術(shù)。警方認(rèn)為，這種欺騙行為可能普遍存在，因?yàn)橹袊?guó)經(jīng)紀(jì)人進(jìn)行了大量指紋修改手術(shù)。這項(xiàng)手術(shù)涉及到摘除拇指和食指上的指紋，然后把它們嫁接到另一只手的手指上。 doi:10.1016/j.ajhg.2011.07.004 PMC: PMID: A Mutation in a Skin-Specific Isoform of SMARCAD1 Causes Autosomal-Dominant Adermatoglyphia Janna Nousbeck, Bettina Burger, Dana Fuchs-Telem, Mor Pavlovsky, Shlomit Fenig, Ofer Sarig, Peter Itin,and Eli Sprecher, Monogenic disorders offer unique opportunities for researchers to shed light upon fundamental physiological processes in humans. We investigated a large family affected with autosomal-dominant adermatoglyphia (absence of fingerprints) also known as the “immigration delay disease.” Using linkage and haplotype analyses, we mapped the disease phenotype to 4q22. One of the genes located in this interval is SMARCAD1, a member of the SNF subfamily of the helicase protein superfamily. We demonstrated the existence of a short isoform of SMARCAD1 exclusively expressed in the skin. Sequencing of all SMARCAD1 coding and noncoding exons revealed a heterozygous transversion predicted to disrupt a conserved donor splice site adjacent to the 3′ end of a noncoding exon uniquely present in the skin-specific short isoform of the gene. This mutation segregated with the disease phenotype throughout the entire family. Using a minigene system, we found that this mutation causes aberrant splicing, resulting in decreased stability of the short RNA isoform as predicted by computational analysis and shown by RT-PCR. Taken together, the present findings implicate a skin-specific isoform of SMARCAD1 in the regulation of dermatoglyph development. |
