Integration of External Signaling Pathways with the Core Transcriptional Network in Embryonic Stem Cells Transcription factors (TFs) and their specific interactions with targets are crucial for specifying gene-expression programs. To gain insights into the transcriptional regulatory networks in embryonic stem (ES) cells, we use chromatin immunoprecipitation coupled with ultra-high-throughput DNA sequencing (ChIP-seq) to map the locations of 13 sequence-specific TFs (Nanog, Oct4, STAT3, Smad1, Sox2, Zfx, c-Myc, n-Myc, Klf4, Esrrb, Tcfcp2l1, E2f1, and CTCF) and 2 transcription regulators (p300 and Suz12). These factors are known to play different roles in ES-cell biology as components of the LIF and BMP signaling pathways, self-renewal regulators, and key reprogramming factors. Our study provides insights into the integration of the signaling pathways into the ES-cell-specific transcription circuitries. Intriguingly, we find specific genomic regions extensively targeted by different TFs. Collectively, the comprehensive mapping of TF-binding sites identifies important features of the transcriptional regulatory networks that define ES-cell identity. |

胚胎干細(xì)胞的外部信號(hào)通路與內(nèi)部轉(zhuǎn)錄網(wǎng)絡(luò)的整合 轉(zhuǎn)錄因子(TFs)及其與靶基因之間的特異性相互作用是說(shuō)明基因表達(dá)程序的關(guān)鍵。為了深入理解ES細(xì)胞的轉(zhuǎn)錄調(diào)節(jié)網(wǎng)絡(luò)，我們用染色質(zhì)免疫沉淀法和超高通量DNA測(cè)序(ChIP-seq)來(lái)呈現(xiàn)13種TFs (Nanog, Oct4, STAT3, Smad1, Sox2, Zfx, c-Myc, n-Myc, Klf4, Esrrb, Tcfcp2l1, E2f1, and CTCF)和2種轉(zhuǎn)錄調(diào)節(jié)因子(p300 and Suz12)的位置。已知這些因子在胚胎干細(xì)胞生物學(xué)中起不同的作用，包括LIF和BMP信號(hào)通路的組成部分、自我更新因子和關(guān)鍵的重新編程因子。我們的研究提供了如下理解：將信號(hào)通路整合到ES細(xì)胞特異性轉(zhuǎn)錄電路中去。有趣的是，我們發(fā)現(xiàn)不同的TFs廣泛地針對(duì)特定基因組區(qū)域�？偟膩�(lái)說(shuō)，TF結(jié)合位點(diǎn)的全面呈現(xiàn)確定了決定ES細(xì)胞種類(lèi)的轉(zhuǎn)錄調(diào)節(jié)網(wǎng)絡(luò)的重要特點(diǎn)。 |
