| 本實(shí)驗(yàn)通過(guò)對(duì)四川各地土樣的采集,用于ε-多聚賴氨酸(ε-poly-lysine,ε-PL)產(chǎn)生菌種的篩選與鑒定。實(shí)驗(yàn)中首先對(duì)采集回來(lái)的泥土用SG液體培養(yǎng)基進(jìn)行富集培養(yǎng)48h,再根據(jù)ε-PL帶正電荷并能與帶正電荷的亞甲基藍(lán)排斥形成透明圈的性質(zhì),通過(guò)加入亞甲基藍(lán)(0.02g/L)和重鉻酸鉀(0.05g/L,作為抑菌劑)的SG固體培養(yǎng)基對(duì)ε-PL產(chǎn)生菌進(jìn)行初篩。其次,根據(jù)ε-PL是一種生物堿,就可以通過(guò)改良碘化鉍試劑鉀(Dragendorff)這種生物堿的顯色劑能與DR試劑形成磚紅色沉淀和甲基橙的磺酸基陰離子能與聚賴氨酸的氨基陽(yáng)離子能形成陽(yáng)離子對(duì),在pH3~10的范圍內(nèi)能形成磚紅色沉淀的原理,通過(guò)對(duì)發(fā)酵上清液進(jìn)行顯色復(fù)篩。然后,再通過(guò)對(duì)篩選出的ε-PL產(chǎn)生菌進(jìn)行生長(zhǎng)特征、形態(tài)特征、生理生化鑒定初步鑒定篩選出的菌屬,再通過(guò)16SrDNA的分子生物學(xué)鑒定方法最終確定出篩選出的菌種種類。 |

| The soil samples collectedaround Sichuan in this experiment were used to screen and identify the production of microbial strain for the ε-poly-L-lysine (ε-poly-lysine,ε-PL) . Then, by the identification of the growth characteristics,morphological and physiological and biochemicalof for the preliminary identification and screening out the production of microbial strain for the ε-PL, and the molecular biology identification of 16SrDNA , the bacteria were screened out ultimately . |
