Also, Fas (fatty acid synthase) or aP2 (adipocyte fatty-acid-binding protein 2), which are both expressed in adipocytes, are known to be involved in insulin resistance.
Fas is expressed in an early stage of adipocyte differentiation and functions to catalyze the synthesis of palmitate from acetyl-CoA and malonyl-CoA. Fas plays a role in the storage of surplus energy in the form of triglycerides, which may cause obesity. The resulting triglyceride palmitate destroys pancreatic β cells to induce insulin resistance。
To examine whether the compounds of Chemical Formulas 1 and 2, which are the active ingredients for improving obesity, can be used as anti-insulin resistance agents, they were applied to adipocytes which were then quantitatively analyzed for resistin, a lipocytokine involved in insulin resistance, and Fas and aP2.
To examine whether the samples are effective as anti-insulin resistance agents, expression levels of resistin, aP2 and Fas in the cells that had been differentiated for 8 days as in Experimental Example 1 were analyzed using the primers of Table 2, below.

另外，脂肪細(xì)胞表達(dá)的脂肪酸合成酶(FAS)或脂肪細(xì)胞脂肪酸結(jié)合蛋白2(AP2)參與胰島素抵抗。
FAS在分化早期階段脂肪細(xì)胞中表達(dá)，其功能是催化乙酰輔酶A和丙二酰輔酶A合成棕櫚酸酯。FAS還促進(jìn)多余的能量以甘油三酯的形式儲(chǔ)存，這可能會(huì)導(dǎo)致肥胖。隨之產(chǎn)生的棕櫚酸甘油酯破壞胰島β細(xì)胞誘導(dǎo)胰島素抵抗。
為了檢測(cè)符合化學(xué)公式1和2的化合物（改善肥胖有效成分）是否可作為抗胰島素抵抗藥物，應(yīng)用它們作用脂肪細(xì)胞，然后定量分析他們對(duì)抵抗素（涉及胰島素抵抗的脂肪細(xì)胞因子）、FAS和AP2表達(dá)的影響。
為了檢測(cè)樣品和抗胰島素抵抗劑是否具有同樣效果，按實(shí)驗(yàn)例1中方法分化細(xì)胞8天，然后應(yīng)用下面表2中引物對(duì)細(xì)胞中抵抗素、AP2和FAS表達(dá)水平進(jìn)行分析。
