More than 34 million people worldwide are currently infected with the human immunodeficiency virus (HIV), the virus that causes AIDS. Yet, how the immune system recognizes and responds to this formidable pathogen is still not clear. In this special feature, we highlight our current understanding of the immune response to HIV with a collection of review and perspective articles. Iwasaki (pages 389–398) summarizes our knowledge of how the innate immune system recognizes HIV and other retroviruses. Bieniasz and colleagues (pages 399–411) cover intrinsic cellular defenses, such as restriction factors. Kwong and Mascola (pages 412–425) highlight what we currently understand about naturally arising neutralizing antibodies to HIV, and Goulder and Walker (pages 426–440) address the relationship between HLA and the virus. Finally, Eisele and Siliciano (pages 377–388) provide a perspective on host reservoirs for HIV and how these represent a challenge to clinicians hoping to eradicate the virus from their patients.
