關于Rat cyclic adenosine monophosphate(cAMP) Elisa Kit 的幾個問題： （1）關于裂解那里我還是沒理解下面這段話一些部分。個人覺得裂解方法是2種： 第一種，直接棄去細胞培養(yǎng)液，加入0.1 M HCl ，孵育10min 或20min，至裂解，（但是加入體積是多少呢？難道加1ml 的0.1 M HCl 那么多嗎？還是我的理解有誤，應該先棄含藥物預處理的培養(yǎng)液，再加入1ml 不含血清的DMEM培養(yǎng)基，其中再加10 ul concentrated hydrochloric acid，這是飽和鹽酸的意思，與0.1 M HCl 是不一樣的吧？），然后離心（600 x g，室溫，多久？），取上清用于接下來elisa。 第二種：先棄含藥物的培養(yǎng)液，然后再加入1ml 的不含血清的DMEM培養(yǎng)基，其中再加 0.1% to 1% Triton x-100（體積呢？） 和 0.1M HCl （10 ul）concentrated hydrochloric acid，然后再離心（600 x g，室溫，多久？），取上清用于接下來的elisa。 關于加了tritonx-100的這種裂解方法，在做elisa會增加光密度值，是不是說在用標準物稀釋成250 pmol/ml, 50 pmol/ml, 10 pmol/ml, 2 pmol/ml, 0.4 pmol/ml, 0.08 pmol/ml時，都是用triton和HCl? 這樣的話，又是怎么配置？相應的NSB和B0孔在第二步加HCL 改為加Triton 和HCL 的混合液。 Cells grown in tissue culture media can be treated with 0.1M HCl after first removing the media. Incubate for 10 minutes and visually inspect the cells to verify cell lysis. If adequate lysis has not occurred incubate for a further 10 minutes and inspect. Centrifuge at 600 x g at room temperature, then use the supernatant directly in the assay. Cell or tissue lysis can be enhanced by adding 0.1% to 1% Triton x-100 to the 0.1M HCl prior to use. When used in this concentration range, the detergent will not interfere with the binding portion of the assay, however there will be a modest increase in the optical density. Samples containing Triton should be evaluated against a standard curve diluted in the same for the most accurate determination. Cyclic AMP in the media can be measured after treating 1 mL of the supernatant media with 10μL of concentrated hydrochloric acid. Centrifuge at 600 x g at room temperature. The supernatants can then be used directly in the assay. 你能不能把你們的做細胞的從裂解到elisa分析的方案發(fā)一份給我。主要是關于裂解加了Triton 之后的。 第二個：關于這個elisa，是基于競爭結合法,我不是很確定，包被的羊-抗鼠的IgG，cAMP, HRP-conjugate,抗體（應該是抗cAMP嗎？）之間是如何結合，比如說是IgG 是競爭結合cAMP 的抗體嗎？為什么它可以結合這種抗體呢?是基于羊與鼠源嗎？cAMP與抗體形成復合物后，HRP-conjugate再與這個復合物結合，這個復合物如何固定在孔板上的呢？洗板主要是洗去多余的游離的HRP-conjugate？ |
