| 阿維菌素發(fā)酵生產(chǎn)過(guò)程中,伴隨著濃重的氣味物質(zhì)—土味素(geosmin)產(chǎn)生,這不僅影響了阿維菌素本身的發(fā)酵代謝過(guò)程,而且降低了目標(biāo)產(chǎn)物的合成效率,更嚴(yán)重的是對(duì)環(huán)境和人類日常生活產(chǎn)生極大的危害。然而,其代謝機(jī)制卻很難單獨(dú)從現(xiàn)有的基因?qū)W、酶學(xué)和發(fā)酵環(huán)境的理論角度進(jìn)行闡明。本課題利用13C同位素標(biāo)記技術(shù)研究土味素在阿維鏈霉菌中與阿維菌素生物合成之間的協(xié)同代謝機(jī)制。首先通過(guò)以特定標(biāo)記的土味素代謝試驗(yàn),探索菌體生長(zhǎng)期和產(chǎn)物合成期土味素在胞內(nèi)的分解代謝機(jī)制和代謝流向;其次,根據(jù)不同13C 標(biāo)記的葡萄糖和土味素組合實(shí)驗(yàn),探索土味素對(duì)生產(chǎn)菌呼吸代謝、中心碳代謝和阿維菌素合成代謝途徑通量變化,揭示阿維鏈霉菌中土味素的合成機(jī)制和代謝網(wǎng)絡(luò)調(diào)節(jié)機(jī)制,實(shí)現(xiàn)其宏觀代謝和微觀代謝研究相結(jié)合,對(duì)阿維菌素發(fā)酵代謝調(diào)控及工藝優(yōu)化有著重要的理論和現(xiàn)實(shí)意義。 |

During the fermentation process of Avermectin, one substance “geosmin” with a strong odor was generated, which will not only affect the fermentation metabolism process itself, but also reduce the synthesis efficiency for the target product. More seriously, it will do a great harm to the environment and everyday life of our human beings. However, it is difficult to elucidate the metabolic mechanism separately from the theoretical point of view of genomics, enzymology and fermentation environment. In this paper, the collaborative metabolism for geosmin between the avermitilis and biosynthesis of avermectin has been investigated by using 13C isotopic labeling technique. Firstly, through the testing metabolic process of geosmin with specific marker, the mechanism of geosmin intracellular catabolism and metabolic flows in the cells during the period of bacterial growth and product synthesis has been explored. Secondly, based on the combination experiments of different 13C-labeled glucose and geosmin, the geosmin-producing bacteria respiration metabolism, central carbon metabolism and pathway flux of avermectin synthesis has been investigated. In this way, the synthesis mechanisms and metabolic network avermitilis adjustment mechanism for geosmin was achieved, resulting in the combination of macro metabolic and micro metabolism, which has important theoretical and practical significance to regulation and optimization the fermentation metabolic process of avermectin. 非本專業(yè),純屬娛樂(lè)。 |
