叉頭轉(zhuǎn)錄因子O亞家族1(forkhead transcription factor group O1,F(xiàn)oxO1)是前體脂肪細(xì)胞分化過程的重要調(diào)控因子,通過多條通路影響細(xì)胞增殖、分化、凋亡但其作用機制及信號通路在魚類的研究中尚不清楚。本項目擬以我國重要經(jīng)濟(jì)魚類一草魚為實驗材料,采用細(xì)胞培養(yǎng)、RT-PCR、實時定量PCR、載體構(gòu)建、免疫組化、細(xì)胞轉(zhuǎn)染、Western blotting及免疫共沉淀結(jié)合質(zhì)譜鑒定技術(shù)等技術(shù)手段,以草魚脂肪細(xì)胞為體外模型,以FoxO1調(diào)控轉(zhuǎn)錄因子PPARγ的活性、RES和Wortmannin阻斷劑對FoxO1的影響及磷酸化FoxO1的亞細(xì)胞定位和作用為切入點,研究FoxO1基因?qū)Σ蒴~脂肪細(xì)胞增殖分化的調(diào)控作用及信號通路。研究結(jié)果不僅能為闡明脂質(zhì)沉積的復(fù)雜網(wǎng)絡(luò)調(diào)節(jié)機制奠定基礎(chǔ),而且還可改為善魚肉質(zhì)量與風(fēng)味提供依據(jù),具有重強的理論及應(yīng)用意義。 [ Last edited by canfeng2008 on 2013-3-7 at 08:10 ] |

Great priorities of the research were given to the control of the activity of transcription actor PPARγ by FoxO1,the influence of RES and Wortmannin blockers on FoxO1 and the locallization and function of phosphorylated FoxO1 subcell ,in order to investigate on the modulation and signalpathways of the proliferation and differentiation of grass carp cell by FoxO1. 研究結(jié)果不僅能為闡明脂質(zhì)沉積的復(fù)雜網(wǎng)絡(luò)調(diào)節(jié)機制奠定基礎(chǔ),而且還可改為善魚肉質(zhì)量與風(fēng)味提供依據(jù),具有重強的理論及應(yīng)用意義。 翻譯:The research result will not only lay the foundation for the clarification of the regulatory mechanism of the complex network of lipid storage,but also provide the basis for improving the flavor and quality of fish,and it is of great significantce theoretically and practically |
