標(biāo)題:針灸對(duì)截癱兔損傷脊髓神經(jīng)再生營(yíng)養(yǎng)修復(fù)影響的機(jī)理研究 中文摘要:目前未見(jiàn)兔脊髓神經(jīng)元的體外培養(yǎng)成功的報(bào)導(dǎo).我們參考鼠神經(jīng)元原代培養(yǎng)的方法,培養(yǎng)兔脊髓神經(jīng)元,作為針灸治療損傷脊髓神經(jīng)元營(yíng)養(yǎng)修復(fù)的主要研究方法。 在既往的研究中,我們指出了針灸對(duì)截癱損傷段的作用可能是:a.促使加速神經(jīng)纖維再生;b.防止瘢痕形成以使神經(jīng)纖維順利通過(guò);c.促使休眠的神經(jīng)元早日蘇醒、受損傷的神經(jīng)元的核蛋白合成等有關(guān)。 本篇研究通過(guò)針灸截癱兔后對(duì)脊髓神經(jīng)元用NGF,bfgf,MMP進(jìn)行培養(yǎng),從細(xì)胞凋亡和凋亡途徑相關(guān)因子FADD、caspase-8、cytochrom C、caspase-9、 caspas-3的表達(dá)差異,細(xì)胞信號(hào)轉(zhuǎn)導(dǎo)途徑的差異(包括H3Tdr測(cè)量、Ca2+濃度、細(xì)胞活力即MTT分析光量度OD值、胞體軸突胞體直徑),鐵超載與轉(zhuǎn)運(yùn)包括(鐵含量、AQP4表達(dá))、脊髓氧化損傷(MDA/Cu.Zn.SOD)與鐵轉(zhuǎn)運(yùn)蛋白等變化差異, 探討針灸對(duì)損傷脊髓神經(jīng)元再生營(yíng)養(yǎng)修復(fù)的機(jī)理。 中文關(guān)鍵詞:1.針灸治療截癱兔;2.脊髓神經(jīng)再生營(yíng)養(yǎng)修復(fù);3.細(xì)胞凋亡;4.細(xì)胞信號(hào)傳遞;5.鐵超載與轉(zhuǎn)運(yùn). The research is mechine about acupuncture and moxibustion to paralysed rabbits on nerve regeneration influcnce of nutritional rehabilitation after spinal cord injury. It is now ,No rabbit spinal cord neurons in vitro success reports , we refer to Big mouse neurons in primary culture method, cultured rabbit spinal cord neurons, as acupuncture treatment spinal cord injury neurons nutritional rehabilitation main research methods In previous studies, we pointed out acupuncture to paraplegia damaged section role may be: a. to promote accelerated nerve fiber regeneration; b. to prevent scar formation in the nerve fibers pass; c. promote an early wake up dormant neurons, promote by damage neuronal in nuclear protein synthesis This study is acupuncture to paraplegia rabbits spinal cord neurons with NGF, bfgf, MMP were cultured, on apoptosis and apoptosis-related factors FADD, caspase-8, cytochrom C, caspase-9, caspas-3 expression differences , cell signal transduction pathways differ (including H3Tdr measurement, Ca2 + concentration, cell viability that MTT assay optical measuring OD value, the cell body axon cell body diameter), iron overload and transportation, including (iron content, AQP4 expression), spinal cord oxidative damage (MDA / Cu.Zn.SOD) and Iron transport protein changes differences , Discussion the mechanism of acupuncture to spinal cord injury neuronal regeneration and nutritional rehabilitation.
