不同性別家蠶【bombyx mori】34℃高溫處理后的基因表達(dá)差異分析【Serial Analysis of Gene Expression(SAGE)】 為了研究家蠶在34℃環(huán)境下耐受高溫的分子作用機(jī)制,我們構(gòu)建了以抗高溫家蠶7532品種5齡幼蟲(chóng)高溫處理前后中腸、絲腺和脂肪體(混合)組織為材料的四個(gè)SGAE文庫(kù)【Serial Analysis of Gene Expression(SAGE)】(常溫(26℃)雌、雄,高溫(34℃)雌、雄)。結(jié)果在獲得的4個(gè)SAGE文庫(kù)中Distinct Tag number為45972、56200、49467、46819,其中能夠比對(duì)到基因的分別占到41.10%、35.00%、35.98%、33.16%,獲得了豐富的數(shù)據(jù)。高溫處理后雌、雄家蠶的基因表達(dá)差異較大,雌蠶高溫后有193個(gè)基因表現(xiàn)上調(diào),507個(gè)基因下調(diào),雄蠶高溫后上調(diào)、下調(diào)基因數(shù)分別為516、318,另外有許多基因在高溫后表現(xiàn)出特異表達(dá)或沉默。差異基因的GO分析(Gene ontology)發(fā)現(xiàn)雌、雄蠶高溫處理后其功能分布基本一致,而在synapse part、virion part、metallochaperone、cell killing、immune system process、rhythmic process顯示出顯著差異(p<0.05)。KEGG (Kyoto Encyclopedia of Genes and Genomes ) pathway分析顯示主要的差異途徑及差異基因個(gè)數(shù)為Oxidative phosphorylation:16個(gè),Purine metabolism:13個(gè),Pyrimidine metabolism:9個(gè),Ubiquitin mediated proteolysis:9個(gè),Aminoacyl-tRNA biosynthesis:8個(gè),Glycolysis / Gluconeogenesis:8個(gè),Arginine and proline metabolism 8個(gè),Ribosome 8個(gè),雖然pathway路徑分析顯示雌雄差異相同,但是每個(gè)途徑中參與的基因種類、基因個(gè)數(shù)及其上下調(diào)模式均表現(xiàn)出較大差異。結(jié)果表明,高溫導(dǎo)致家蠶基因表達(dá)水平上的明顯差異,而這些差異基因也顯示出了性別差異,這些數(shù)據(jù)可以為研究不同性別家蠶對(duì)高溫耐受的分子機(jī)理提供研究基礎(chǔ)。 |

An Serial Analysis of Gene Expression(SAGE)of Bombyx Moris with Different Sex in 34℃ To study the molecular mechanisms of temperature-resistance of bombyx mori in 34℃, this experiment has used the middle intestines, silk glands and fat bodies of the 5th instar larvae of Temperature-resistance Bombyx Mori Specie No. 7532 before and after heat treatment as materials to construct four Serial Analysis of Gene Expression(SAGE)libraries, namely, male and female in normal temperature of 26℃, male and female in high temperature of 34℃.The results indicate that the Distinct Tag Numbers of the four SAGE libraries are 45972、56200、49467、46819 respectively, and the rates of mapping to genes are 41.10%、35.00%、35.98%、33.16% in the four libraires accordingly. Abundant data are obtained through the experiment.There are large differences between the generic expressions of male and female Bombyx Moris after heat treatment. 193 genes of female Bombyx Mori are up-regulated and 507 genes are down-regulated; while the up-regulated number and down-regulated number of genes of male Bombyx Mori are 516 and 318 respectively. Additionally, plenty of genes are expressed specifically or being silenced after heat treatment.The gene ontology (GO) analysis of differential genes claims that the functional distributions of male and female Bombyx Mori after heat treatment are roughly identical, however, significant differences(p<0.05) are showed in synapse part, virion part, metallochaperone, cell killing, immune system process and rhythmic process. The KEGG (Kyoto Encyclopedia of Genes and Genomes ) Pathway Analysis reveals the major distinct ways and the number of distinct genes: Oxidative phosphorylatio: 16, Purine metabolism:13, Pyrimidine metabolism: 9, Ubiquitin mediated proteolysis: 9, Aminoacyl-tRNA biosynthesis: 8, Glycolysis / Gluconeogenesis: 8, Arginine and proline metabolism: 8, Ribosome: 8. Although the distinctions of male and female bombyx moris are shown to be similar in the pathway analysis, the sorts, numbers and up/down-regulated models of genes in every single way vary considerably. The results show that high temperature has caused significant differences in generic expression levels of bombyx mori , and those distinct genes reveals gender differences.
