DLL4 is a ligand for the Notch family of receptors. DLL4 has many important functions in normal development and tissue homeostasis, including roles in the immune system, the gastro-intestinal tract, and in vascular development. Because of the importance of Notch signaling in stem cell biology, DLL4 has been investigated for its role in the maintenance and proliferation of cancer stem cells (CSC) . In addition, its important role in angiogenesis has been investigate d for utility as an anti-angiogenic agent. Preclinical studies have highlighted that both anti-CSC and anti-angiogenic activities contribute to its anti-tumor efficacy, and have supported the clinical development of anti-DLL4 antibody for the treatment of cancer |

DII4是一種內(nèi)皮特異性配體，其通常在血管發(fā)育及血管生成部位表達(dá)。已經(jīng)證實(shí)，在乳腺癌、腎癌、膀胱癌的血管分布中，DII4的表達(dá)會(huì)明顯上調(diào)。 DII4是NOTCH家族受體的配體。DII4在正常發(fā)育和組織體內(nèi)平衡如免疫系統(tǒng)、胃腸道及血管生成等方面具有重要作用。 由于在干細(xì)胞生物學(xué)中，Notch信號(hào)有非常重要的作用，因此已有許多研究對(duì)DLL4在癌癥干細(xì)胞增殖及維持方面的作用進(jìn)行了考察。 臨床前研究顯著證實(shí)抗CSC與抗血管生成活性均有抗腫瘤效果，而且這一研究對(duì)抗DLL4抗體在癌癥治療方面的臨床發(fā)展具有重要的支持作用。 |
