| 抗菌肽（Antimicrobial peptides）是一類(lèi)具有抗菌活性的短肽，分子量在1000-7000左右，由12-50個(gè)氨基酸組成，具有光譜抗菌性，可殺死革蘭氏陽(yáng)性菌、革蘭氏陰性菌、真菌等。抗菌肽ORBK是來(lái)源于無(wú)指盤(pán)臭娃Odorrcma grahami皮膚分泌物的含16個(gè)氨基酸殘基的陽(yáng)離子多肽。該多肽是一類(lèi)環(huán)形肽，不僅具有抗菌活性，還具有穩(wěn)定的促胰島素釋放活性。目前獲得抗菌肽ORBK的方法只有提取和合成，從無(wú)指盤(pán)臭娃皮膚分泌物中提取存在提取量少的瓶頸，而化學(xué)合成法成本較高，因?yàn)樵摱嚯暮铣芍羞�需要一個(gè)環(huán)化的過(guò)程。在本論文中，我們利用基因工程結(jié)合H2O2氧化的方法，成功獲得了抗菌肽ORBK，并對(duì)該抗菌肽的作用機(jī)制進(jìn)行了探究，同時(shí)，我們還對(duì)抗結(jié)合桿菌多肽D29P63和PK34進(jìn)行了初步研究。 |

Antimicrobial peptides are antimicrobial peptides with molecular weight of about 1000-7000 and consists of 12-50 amino acids whose antibacterial spectrum include Gram-positive bacteria, Gram-negative bacteria and fungi. ORBK antimicrobial peptides are cationic peptides derived from Odorrcma grahami skin secretions containing 16 amino acid residues. A cyclic peptide, ORBK antimicrobial peptides have not only antimicrobial activity , but also a stable insulinotropic activity. Currently ORBK antimicrobial peptide can only be extracted and synthesized . The much smaller yields of Odorrcma grahami skin secretions extraction is a critical technical bottleneck and the cost of chemical synthesis is high because it requires a cyclic process . In this paper, we use a genetic engineering combined H2O2 oxidation method, successfully obtained ORBK antimicrobial peptides .The mechanism of antimicrobial peptides has been explored and bacteria binding peptides D29P63 and PK34 were studied also . 無(wú)指盤(pán)臭娃Odorrcma grahami 請(qǐng)核對(duì)。 |
