本論文的工作主要包括三部分: 1. 抗菌肽ORBK的制備及分子機(jī)制研究。利用基因工程法把抗菌肽ORBK與融合蛋白MBP融合表達(dá)純化,酶切后通過HPLC方法分離獲得線性與環(huán)化混合抗菌肽ORBK,通過H2O2氧化方法獲得環(huán)化抗菌肽ORBK。 2. 大多數(shù)抗菌肽的作用機(jī)制是破壞細(xì)胞膜的結(jié)構(gòu),引起細(xì)胞內(nèi)容物大量滲透,最終導(dǎo)致細(xì)菌死亡。我們利用Nanodiscs和liposome體系對抗菌肽ORBK的作用機(jī)制進(jìn)行了研究,研究表明抗菌肽ORBK作用于細(xì)菌細(xì)胞膜,并導(dǎo)致細(xì)菌細(xì)胞內(nèi)容物泄漏,從而導(dǎo)致細(xì)菌死亡。 3. D29P63和PK34是D29分枝桿菌噬菌體來源的多肽。它們可作為制備抗結(jié)核桿菌藥物制劑的應(yīng)用,我們利用基因工程法成功獲得了這兩種多肽,并首次利用液體核磁共振技術(shù)完成了它們的主鏈原子化學(xué)位移的指認(rèn)工作。 |

This paper mainly focus on the following three parts: 1 Preparation and molecular mechanisms of ORBK antimicrobial peptides . Realized the ORBK antimicrobial peptide fusion protein MBP's fusion expression and purification using genetic engineering method and obtained cyclic ORBK antimicrobial peptides through separation of the mixed linear and cyclic ORBK antimicrobial peptides by HPLC after digestion method and H2O2 oxidation . 2The mechanism of most of antimicrobial peptides is destroy the structure of cell membranes which caused permeation of a large number of cell contents and resulted in bacterial death. We used liposome system and Nanodiscs and to study the mechanism of ORBK antimicrobial peptides . Result showed the ORBK antimicrobial peptides's effect on bacterial membrane caused the leakage of the contents and resulted in bacterial death. 3. D29P63 and PK34 are D29 Mycobacteriophage peptides which can be used to prepare anti-TB drugs formulations. We successfully obtained these two peptides by genetic engineering method and for the first time completely identified their main chain atoms by liquid nuclear magnetic resonance technology |
