中文：普拉特霉素是一種大環(huán)內(nèi)酯類抗生素。本文建立了一種利用A將麥迪霉素轉(zhuǎn)化為普拉特霉素的方法，發(fā)酵產(chǎn)量達(dá)到5.528g/L。通過調(diào)節(jié)pH進(jìn)行反復(fù)沉淀溶解粗提取與ODS反相層析分離精提取相結(jié)合的方法對發(fā)酵液中的普拉特霉素A1進(jìn)行分離純化，得到純度為97.74%的純品，對所得純品進(jìn)行了MALDI-TOF-MS和NMR檢測，確定了所得物質(zhì)為普拉特霉素。對提純后的普拉特霉素A1進(jìn)行了體外藥效試驗(yàn)，驗(yàn)證了普拉特霉素對Staphyloccocus aureu、Streptococcus pneumoniae和Septic Streptococcus和Porcine Actinobacillus pleuropneumoniae都具有較好的藥物敏感性。 英文：Platenomycin is a macrolide antibiotic. The process for producing antibiotic composition Platenomycin by bioconversion of Midecamycin by A was investigated. The highest yield of Platenomycin was 5.654g/L. Recovering the antibiotic from the fermentation broth by adjusting the pH, and purifying the crude residue to 97.74%. The purified compound was identified as Platenomycin by MALDI-TOF-MS and NMR. | The preparation of Platenomycin, a macrolide antibiotic, through bioconversion of Midecamycin using A was investigated, giving the highest yield of Platenomycin at 5.654g/L. A purity of 97.74% was achieved after recovering the antibiotic from the fermentation broth through adjusting pH, and multiple purification of the crude residue. The final compound was verified as Platenomycin by MALDI-TOF-MS and NMR. | Platenomycin is a kind of macrolide antibiotic. By using A,we established a method to turn Midecamycin into Pratt neomycin ,with which fermentation production come to 5.528g / L. Through adjusting the PH to repeatedly precipitation and dissolve the crude extract combined with ODS reversed phase chromatography method ,the Pratt ADM A1 in the fermentation broth were isolated and purified.The product is of 97.74% purity and identified of Pratt neomycin by MALDI-TOF-MS and NMR detection. In vitro efficacy trials conducted of the purified Pratt ADM A1 verified that Pratt ADM has better drug sensitivity on Staphyloccocus aureu,Streptococcus pneumoniae,Septic Streptococcus and Porcine Actinobacillus pleuropneumoniae. |
