| 分別比較化合物1、2與MAO-A、MAO-B的對接模式可知,降碳倍半萜單元的引入造成分子的體積和表面積明顯增大。在與MAO口袋作用時,兩個配體與口袋的作用都是以疏水作用為主,化合物1中的降碳倍半萜單元與額外的疏水腔形成了相互作用,導(dǎo)致配體1與口袋的結(jié)合能力增強。另一方面,較大取代基的引入,口袋對化合物1的擠壓,使化合物1整體向FAD靠近,因而造成它與FAD的結(jié)合更為緊密。因此會在實驗上發(fā)現(xiàn)化合物1接入降碳倍半萜單元后活性提高了幾倍。這一點從下表對接的結(jié)合自由能也能看出。 |

The comparison of the docking mode between compound 1 or 2 and MAO-A as well as MAO-B showed that with the introduction of carbon reducing sesquiterpene unit,the molecular volume and surface area significantly increased. When they are interacted with MAO pocket, the interaction between the two ligands and pockets are mainly hydrophobic interactions and carbon reducing sesquiterpene unit of compound 1 interacted with the additional sesquiterpene cavity,which lead to the enhancement of combination capacity of compound 1and the pocket. On the other hand, with the introduction of a larger substituent,the pocket will squeeze compound 1 and approch compound 1 as a whole on FAD which caused more closely combination with FAD. That is why activity of compound 1 in experiment increased several times after the introduction of carbon reducing sesquiterpene unit which can also be seen in the binding free energy of docking of the following table. 自己再看看吧。 |
