德國(guó)應(yīng)化Angewandte Chemie International Edition上新發(fā)表一篇關(guān)于材料表面抗蛋白吸附的綜述,并且封面推薦。 Protein Interactions with Polymer Coatings and Biomaterials DOI: 10.1002/anie.201400546 正文 http://onlinelibrary.wiley.com/d ... .201400546/abstract 封面 http://onlinelibrary.wiley.com/d ... .201406350/abstract 此文為Freie Universitt Berlin,Helmholtz-Zentrum Berlin,Helmholtz-Zentrum Berlin中三個(gè)相關(guān)領(lǐng)域大組合寫 小妹做此領(lǐng)域工作,剛好讀到這篇文章,覺得這篇綜述很獨(dú)到,不僅僅綜述了材料方面新進(jìn)展,還從機(jī)理,模擬,分析表征,應(yīng)用等多方位探討蛋白吸附問題,在此愿將好文與關(guān)注此領(lǐng)域的蟲友一起分享。 此文第一部分Introduction,主要介紹蛋白吸附機(jī)理,其中提到了Whitesides rules和Bergs law的局限性與新發(fā)展,并對(duì)指導(dǎo)抗蛋白吸附材料設(shè)計(jì)的Whitesides rules進(jìn)行了新詮釋。 第二部分Methods for the Analysis of Protein Interactions,介紹了目前表征蛋白吸附的常用方法,對(duì)一些新方法QCM、SPR、ToF-SIMS、ITC、FTIR等進(jìn)行了詳細(xì)介紹,此外詳述了生化表征手段和計(jì)算機(jī)模擬方法。 第三部分Bioinert and Biospecific Surfaces-Recent Developments and Applications,著重介紹抗蛋白吸附材料的研究新進(jìn)展及其應(yīng)用。不同于其他大部分此領(lǐng)域綜述按材料分類介紹,此文從coating結(jié)構(gòu)著手,討論了不同coating形式的利弊,并且在第20頁有一段專門的Guidelines for the Design of a Protein-Resistant Surface Coating。 第四部分Conclusions and Future Perspectives,展望了抗蛋白吸附研究中的新問題和發(fā)展方向。有幾個(gè)問題提得很獨(dú)到,比如越來越多的研究設(shè)計(jì)到蛋白構(gòu)象變化,但蛋白構(gòu)象變化對(duì)生物相容性的影響還研究甚少。 摘要 Protein adsorption is considered to be the most important factor of the interaction between polymeric biomaterials and body fluids or tissues. Water-mediated hydrophobic and hydration forces as well as electrostatic interactions are believed to be the major factors of protein adsorption. A systematic analysis of various monolayer systems has resulted in general guidelines, the so-called “Whitesides rules”. These concepts have been successfully applied for designing various proteinresistant surfaces and are being studied to expand the understanding of protein–material interactions beyond existing limitations. Theories on the mechanisms of protein adsorption are constantly being improved due to the fast-developing analytical technologies. This Review is aimed at improving these empirical guidelines with regard to present theoretical and analytical advances. Current analytical methods to test mechanistic hypotheses and theories of protein–surface interactions will be discussed.
