| 結(jié)核病是由人類重大病原菌結(jié)核桿菌引發(fā)的呼吸道傳染病,隨著多種有效控制結(jié)核病的抗生素的大量應(yīng)用,多重耐藥結(jié)核桿菌菌株大量出現(xiàn),而傳統(tǒng)抗生素?zé)o法達到治療效果,因此,開發(fā)新型抗結(jié)核桿菌藥物迫在眉睫。噬菌體作為一種真細菌的病毒,可以有效地與宿主細菌作用,作為替代傳統(tǒng)抗生素的藥物受到廣泛的關(guān)注。多肽PK34來源于結(jié)核分枝桿菌噬菌體,具有清除體內(nèi)結(jié)核分枝桿菌的能力,但是,目前其抗結(jié)核分枝桿菌的作用機制研究還未有報道,本項目擬利用基因工程法獲得抗結(jié)核分枝桿菌多肽PK34,并利用核磁共振為主要研究手段,結(jié)合圓二色譜等其他手段,研究抗結(jié)核分枝桿菌多肽PK34與TDM相互作用的動態(tài)過程及其在TDM存在情況下的溶液結(jié)構(gòu),并探索抗結(jié)核分枝桿菌多肽PK34的抗結(jié)核分枝桿菌作用機理,為闡明其抗結(jié)核分枝桿菌的作用機理提供重要的理論依據(jù)。 |

With the large-scale application of a variety of effective antibiotics (to control TB), multidrug-resistant Mycobacterium tuberculosis strains emerged, which cannot be treated effectively with the traditional antibiotics. Therefore, it is the imminent to develop new anti-TB drugs. As a virus of eubacteria, a bacteriophage can infect and replicate within Mycobacterium tuberculosis. Recently, much attention has been paid to bacteriophage as an alternative to traditional antibiotic drugs. Polypeptide PK34, derived from Mycobacterium tuberculosis phage, is capable of getting rid of Mycobacterium tuberculosis from body. However, the underlying mechanisms of its anti-Mycobacterium tuberculosis effect has not been reported. In this project, polypeptide PK34 of Mycobacterium tuberculosis will be obtained by utilizing of genetic engineering methods and the dynamic interaction between PK34 and TDM, as well as the solution structure of PK34 in the presence of TDM will be investigated with MRI as the main research tool, in combination with other methods, such as circular dichroism. The mechanisms of the anti-Mycobacterium tuberculosis effect of polypeptide PK34 will be explored in order to provide important theoretical basis for its therapeutic action. |
