【摘要】 目的 旨在在HEK293細(xì)胞系和Flp-IN-CHO細(xì)胞中構(gòu)建穩(wěn)定表達(dá)超極化激活環(huán)核苷酸門(mén)控鉀通道亞型4(HCN4)的穩(wěn)定細(xì)胞系,并對(duì)其通道的表達(dá)進(jìn)行鑒定。 方法 利用Lipofectamine2000將pCDNA3.1(+)-HCN4分別轉(zhuǎn)染進(jìn)入HEK293細(xì)胞和Flp-IN-CHO細(xì)胞中,并通過(guò)G418和潮霉素B分別對(duì)其進(jìn)行篩選,獲得陽(yáng)性單克隆細(xì)胞系,采用全細(xì)胞膜片鉗技術(shù)對(duì)篩選出的單克隆細(xì)胞進(jìn)行表達(dá)水平的鑒定,測(cè)定pCDNA3.1(+)-HCN4 所介導(dǎo)的快速激活延遲整流鉀電流(If)。結(jié)果 全細(xì)胞膜片鉗實(shí)驗(yàn)在篩選出的單克隆細(xì)胞中記錄到超極化激活的非選擇內(nèi)向陽(yáng)離子電流(If)。 結(jié)論 該方法有效地將目的基因轉(zhuǎn)染進(jìn)入HEK293細(xì)胞和Flp-IN-CHO細(xì)胞中,并能穩(wěn)定表達(dá)HCN4通道蛋白。HCN4通道基因重組載體的成功構(gòu)建為進(jìn)一步研究該離子通道的電生理功能及在疾病治療方面的潛在應(yīng)用價(jià)值奠定了基礎(chǔ)。![]() ![]() ![]() ![]() |

We then used whole cell membrane clumping technique to assess the level of gene expression by measuring pCDNA3.1(+)-HCN4-dependent 快速激活延遲整流鉀電流 (太專業(yè)了lz自己翻譯吧) channel activities (If) . 結(jié)果 全細(xì)胞膜片鉗實(shí)驗(yàn)在篩選出的單克隆細(xì)胞中記錄到超極化激活的非選擇內(nèi)向陽(yáng)離子電流(If)。 Results Whole cell membrane clumping technique was used successfully to record the ultra polarized activation of non-selective inward cation current on monoclonal stable cell lines. 結(jié)論 該方法有效地將目的基因轉(zhuǎn)染進(jìn)入HEK293細(xì)胞和Flp-IN-CHO細(xì)胞中,并能穩(wěn)定表達(dá)HCN4通道蛋白。HCN4通道基因重組載體的成功構(gòu)建為進(jìn)一步研究該離子通道的電生理功能及在疾病治療方面的潛在應(yīng)用價(jià)值奠定了基礎(chǔ)。 Conclusion: This methodology can be used to introduce target genes into HEK293 and Flp-IN-CHO cells by transfection and stably express HCN4 channel protein. The construction of expression plasmid for HCN4 gene and stable lines pave the road for further electrophysiology studies for its normal function and potential application in treating certain diseases. |
