| 結(jié)果顯示:模型組小鼠肺組織中PI3K、AKT1/2及NF-κBp65蛋白的表達(dá)水平在不同時(shí)間均高于正常組(p<0.05),PI3K、AKT1/2于感染第5天時(shí)表達(dá)最高,NF-κBp65表達(dá)呈持續(xù)增高狀態(tài)。與模型組比較,桑白皮黃酮組小鼠肺組織中PI3K、AKT1/2蛋白的表達(dá)水平在各時(shí)間點(diǎn)顯著降低(p<0.05),NF-κBp65于感染第5、7天時(shí)表達(dá)水平顯著降低(p<0.05);桑白皮多糖組小鼠肺組織中PI3K蛋白的表達(dá)水平于感染第3 、5天時(shí)顯著降低(p<0.05),AKT1/2蛋白的表達(dá)水平于感染第3、7天時(shí)顯著降低(p<0.05),NF-κBp65蛋白的表達(dá)水平各時(shí)間點(diǎn)均顯著降低(p<0.05)。結(jié) 論:1、采用RSV long株經(jīng)鼻腔連續(xù)滴鼻的方法可以成功造出RSV感染幼齡BALB/c小鼠RSV肺炎模型。2、RSV感染小鼠后,可引起小鼠血清IFN-γ、IL-4與肺組織PI3K、AKT1/2及NF-κBp65表達(dá)的改變。3、桑白皮不同有效成分可以調(diào)控RSV肺炎小鼠PI3K/AKT信號(hào)通路與血清IFN-γ、IL-4的表達(dá),其可能是桑白皮防治RSV肺炎、減輕其炎癥反應(yīng)的作用機(jī)制之一。關(guān)鍵詞:呼吸道合胞病毒(RSV);BALB/c小鼠;桑白皮;IFN-γ;IL-4;PI3K/AKT/ NF-κB信號(hào)通路;肺炎 |

| The results showed that the levels of PI3K and AKT1 / 2 and NF-κB p65 protein expression on different time were higher in the lung tissue of mice from the model group than those of the normal group (P < 0.05), the expression levels of PI3K and AKT1 / 2 peaked by day 5, while the NF-κB p65 expression was persistently elevated. Compared with the lung tissues from the model group, those from Cortex mori flavonoids-treated (and infected) mice had significantly reduced PI3K and AKT1 / 2 protein expression levels in all time points (P < 0.05), and the NF-κB p65 levels were significantly reduced (P < 0.05) in lungs from the infected mice on days 3, and 5; In Cortex mori polysaccharides-treated mice group, the lung PI3k expression levels were also significantly decreased (P < 0.05) on days 3 and 5 post infection, the expression levels of AKT1 / 2 were significantly reduced (P < 0.05) on days 3 and 7 post infection and , NF-κB p65 protein expression level were significantly lower (P < 0.05) in all time point. Conclusion: 1, successive intranasal infection with the RSV long strains can successfully generate RSV pneumonia model in young BALB / c mice. 2, RSV infection can trigger changes in the levels of serum IFN-γ and IL-4 mice, and PI3K, AKT1/2 and NF-kB p65 expression levels in lung tissue. 3, the white bark from mulberry root have different active ingredients that can regulate the PI3K, Akt signaling pathways and serum IFN gamma, IL-4 expressions in mice with RSV pneumonia, which may be the mechanism of action for Mulberry bark in prevention and treatment of RSV pneumonia, and alleviating the severity of inflammatory reaction.
