| 溫陽(yáng)方由溫陽(yáng)利水法化裁而來(lái)，是王老經(jīng)長(zhǎng)期臨床實(shí)踐總結(jié)的慢性心衰中藥復(fù)方制劑。前期臨床和實(shí)驗(yàn)研究證實(shí)該方具有良好的臨床療效，能夠改善心力衰竭動(dòng)物模型心肌重塑。隨著慢性心力衰竭研究進(jìn)展的不斷深入，細(xì)胞因子網(wǎng)絡(luò)在心力衰竭心肌重構(gòu)中的作用日益引起廣泛關(guān)注。其中腫瘤壞死因子α(Tumor necrosis factorα，TNFα) 是重要的促炎性細(xì)胞因子，作為激活細(xì)胞級(jí)聯(lián)反應(yīng)的主要介質(zhì)，在細(xì)胞因子網(wǎng)絡(luò)中發(fā)揮關(guān)鍵作用。核轉(zhuǎn)錄因子-κB (nuclear factor κB，NF-κB)是調(diào)控多種基因轉(zhuǎn)錄的關(guān)鍵因子，活化后能調(diào)控包括TNFα在內(nèi)的多種炎癥介質(zhì)的轉(zhuǎn)錄，作為炎性反應(yīng)的整合者在免疫調(diào)節(jié)和炎癥發(fā)生過(guò)程中具有重要作用。為進(jìn)一步探討溫陽(yáng)方藥效和作用機(jī)理，明確NF-κB在心衰發(fā)病中的作用及對(duì)TNFα調(diào)節(jié)機(jī)制，本研究采用細(xì)胞培養(yǎng)和血清藥理學(xué)方法，開(kāi)展溫陽(yáng)方對(duì)腫瘤壞死因子α（TNFα）誘導(dǎo)新生大鼠心肌成纖維細(xì)胞（cardial fibroblasts CFs）增殖、膠原合成，及核轉(zhuǎn)錄因子NF-κB表達(dá)的影響，進(jìn)一步從分子水平探討其抑制心肌纖維化，改善心力衰竭心肌重塑的作用機(jī)制，從而為其臨床應(yīng)用及新藥開(kāi)發(fā)提供科學(xué)理論和依據(jù)。 |

| By the WenYangFang WenYangLi water cut and come, is the long-term clinical practice cenwanglaoshan herbal preparations of chronic heart failure. The clinical and experimental research confirms the party has good curative effect, can improve cardiac remodeling animal model for heart failure. With chronic heart failure, research progress of the cytokine network in hf myocardial reconstruction is aroused widespread concern. Among Tumor necrosis factor alpha (Tumor necrosis, TNF alpha) in alpha is an important factor of inflammatory cells, promote cell activation cascade as the main medium, in cytokines play an important role in network. Transcription factor - κ B (nuclear κ B, NF - in κ B) is the regulation of gene transcription factors, activate the key to control including TNF alpha, inflammation, as the transcription of integration of the inflammatory response in immune adjustment and inflammation plays an important role in the process. For further discussion WenYangFang efficacy and mechanism, clear in heart failure κ B NF - the role and pathogenesis of TNF alpha adjusting mechanism, this study adopts cell culture and serum pharmacology, WenYangFang method of tumor necrosis factor (TNF alpha) inducing newborn rat cardiac fibroblasts cardial (CFs) proliferation, fibroblasts collagen synthesis, and κ transcription factor NF - expression of B, further discusses its suppression of the molecular myocardial fibrosis, improve cardiac remodeling of heart failure mechanism, so as to its clinical application and development of new medicine to provide scientific theory and evidence. |
