Reviewer #1: 1. Introduction. Why do the authors choose a rabbit model? What are the advantages of this animal? Why not a larger animal model, such as a dog or sheep? 2. Methods. What kind of rabbit was used? What diet was it fed on (normal or high cholesterol)? 3. Methods. Did the authors excise a piece of the carotid artery? How long a graft was used? What was its diameter? What sutures were used to suture the vein graft? 4. Results, Figure 1. What was the time until 50% of the drug was eluted? It appears to be approximately 21-28d in Figure 1F, but an exact calculation would be useful. 5. Results, Figure 2. This figure would be more convincing if Panels B and C were aligned in the same direction, and a scale bar was given (showing panels B and C to be higher magnification). 6. Results, Figure 2. How was the intimal thickness calculated? Manually at several spots, or with computer-aided morphometry? Was the thickness based off of an elastin stain for accuracy? 7. Results, Figure 3. The authors use immunohistochemistry as an exclusive method of detection. Did the authors confirm their findings with a Western blot? 8. Do the authors believe that rapamycin will be clinically useful for human patients? 9. Does rapamycin suppress the rabbit immune system? Did the authors measure immune system function? 10.
