多劑量口服10mg兩種制劑,每日一次,每次10mg,于服藥后第4天血藥濃度達穩(wěn)態(tài)。兩種制劑的F值為98.9%±7.9%,參數(shù)Tmax,Cmax,Cmin,AUCss,AUMCss,MRT,F(xiàn)I等主要藥代參數(shù)接近,經(jīng)方差顯示無顯著性差異,進一步經(jīng)雙單側t檢驗和(1-2a)置信區(qū)間分析顯示藥代動力學參數(shù)接受生物等效性假設,說明兩種制劑等效。 臨床觀察,試驗期間未見不良反應,無中途終止試驗的受試者。 體內外相關性 體外溶出數(shù)據(jù) 依據(jù)研制單位制定的釋放度方法測定試驗緩釋膠囊的體外溶出數(shù)據(jù)如表 體內吸收數(shù)據(jù) 以Wanger-Nelson法計算試驗用緩釋膠囊不同時間的體內吸收數(shù)據(jù),結果如表 體內吸收數(shù)據(jù)與體外溶出分數(shù)的相關性良好,相關方程如下: 體內吸收量%=1.455*體外溶出量-12.23 r=0.9638 |

Plasma concentration reached steady state at the fourth day after administration. F value for two preparations was 98.9%±7.9%, ANOVA analysis showed no significant differences between two preparations on Tmax,Cmax,Cmin,AUCss,AUMCss,MRT and FI. Further analysis by two- tailed T-test and (1-2a) confidential interval suggested that pharmacokinetic parameters of two preparations were comparable. 臨床觀察,試驗期間未見不良反應,無中途終止試驗的受試者。 No side effects were found during the trial. The data was shown in table 1. 體內吸收數(shù)據(jù) In vivo absorption study 以Wanger-Nelson法計算試驗用緩釋膠囊不同時間的體內吸收數(shù)據(jù),結果如表 體內吸收數(shù)據(jù)與體外溶出分數(shù)的相關性良好,相關方程如下: 體內吸收量%=1.455*體外溶出量-12.23 r=0.9638 In vivo absorptions in different time were examined by Wanger-Nelson method.
