| 背景與目的:實(shí)驗(yàn)通過建立模擬人皮膚取皮區(qū)創(chuàng)面來檢測生長因子TGF-β1和VEGF在創(chuàng)面愈合以及瘢痕增生方面的作用,為臨床應(yīng)用新型藻酸鹽敷料治療取皮區(qū)創(chuàng)面的應(yīng)用提供數(shù)據(jù)依據(jù),方法:主要將藻酸鹽敷料應(yīng)用于版納微型豬近交系中厚皮取皮區(qū)創(chuàng)面,通過檢測不同時(shí)間點(diǎn)(取皮后0、3、7、14、28天)創(chuàng)面愈合過程中及創(chuàng)面愈合后的生長因子TGF-β1、VEGF的mRNA表達(dá)情況,探討新型藻酸鹽敷料對創(chuàng)面愈合的生物學(xué)機(jī)制及減輕瘢痕增生的具體作用機(jī)制。結(jié)果:構(gòu)建的取皮區(qū)創(chuàng)面動物模型,從大體外觀上觀測,其藻酸鹽治療組與凡士林對照組存在明顯差異,治療組創(chuàng)面愈合時(shí)間少于對照組(P<0.05),具有統(tǒng)計(jì)學(xué)差異。取皮區(qū)創(chuàng)面愈合過程中不同時(shí)間點(diǎn)的治療組與對照組的基因表達(dá)情況表明,不同的時(shí)間點(diǎn)之間有顯著差異(P<0.01),治療與否有顯著性差異(P=0.025),不同豬之間無統(tǒng)計(jì)學(xué)差異。TGF-β1在0、3、7、14、28天的表達(dá)量為:治療組-5.068±0.286,-4.131±0.396,-3.294±0.516,-3.514±0.374,-4.695±0.809;對照組-5.068±0.286,-4,457±0.450,-4.262±0.512,-4.025±0.365,-4.485±0.233。TGF-β1表現(xiàn)為隨著創(chuàng)面的愈合其表達(dá)上升,至中期表達(dá)升至最高,之后表達(dá)下降,在創(chuàng)面完全愈合后其表達(dá)量趨于正常水平。其中在14天藻酸鹽治療組與凡士林對照組之間TGF-β1表達(dá)有顯著差異。VEGF的mRNA表達(dá)量的統(tǒng)計(jì)結(jié)果與TGF-β1的結(jié)果相似,其中7天藻酸鹽治療組與凡士林對照組之間VEGF的mRNA的表達(dá)有顯著差異。VEGF在0、3、7、14、28天的表達(dá)量為:治療組-3.461±0.279,-2.726±0.425,-1.722±0.097,-2.401±0.341,-2.902±0.437,對照組為-2.928±0.385,-2.522±0.319,-2.877±0.517,-3.230±0.451。結(jié)論:通過構(gòu)建取皮后中厚皮創(chuàng)面動物模型,成功構(gòu)建了檢測取皮區(qū)創(chuàng)面組織中VEGF、TGF-β1的mRNA的SYBR GreenⅠ實(shí)時(shí)熒光定量檢測方法,揭示藻酸鹽敷料對促進(jìn)創(chuàng)面愈合有重要作用,且主要與調(diào)控細(xì)胞因子的表達(dá)相關(guān),為解決創(chuàng)面愈合減少瘢痕增生等臨床棘手問題提供了可靠的理論依據(jù)。 |

Background and Objective:we detect the influence of growth factor TGF-β1 and VEGF in wound healing and scar formation through the establishment of simulation of people skin in access site wound. it provides the data for clinical application of new alginate dressings on skin treatment Wounds . Method:we mainly applied alginate dressing in inbred Banna minipig check site wounds in the thick. we detect the different time points (0,3,7,14,28 days after taking the skin) and wound healing process of wound healing growth factor TGF-β1, VEGF expression of the mRNA to explore new alginate dressing on wound healing biological mechanisms and the specific role of reducing scar formation mechanism. 吃完中飯回來繼續(xù)翻 ![]() |
| Results: we observe from the general appearance of construction of access site wound model, , the Vaseline alginate treatment group and control group was significantly different, healing time of the treatment group was shorter than the control group (P <0.05), which is significantly different. Skin graft taken at different time points during the treatment and control groups showed that the gene expression of the different time points were significantly different(P <0.01), whether the treatment significantly different or not (P = 0.025), no statistical difference between the different pigs exists. |
TGF-β1 expression in 0,3,7,14,28 days as follows:Treatment group, -5.068 ± 0.286, -4.131 ± 0.396, -3.294 ± 0.516, -3.514 ± 0.374, -4.695 ± 0.809; control group, -5.068 ± 0.286, -4,457 ± 0.450, -4.262 ± 0.512, -4.025 ± 0.365, - 4.485 ± 0.233.As TGF-β1 showed increased the expression of wound healing, increased expression of the highest to medium term, after which expression decreased in the wound healed completely after its expression tends to the normal level.Alginate treatment group and control group Vaseline expression of TGF-β1 were significantly different in 14 days.The amount of VEGF mRNA expression was similar to TGF-β1 , however,expression of VEGF mRNA in alginate treatment group and control group Vaseline were significantly different.VEGF expression in 0,3,7,14,28 days as follows: the treatment group -3.461 ± 0.279, -2.726 ± 0.425, -1.722 ± 0.097, -2.401 ± 0.341, -2.902 ± 0.437, control group was -2.928 ± 0.385, -2.522 ± 0.319, -2.877 ± 0.517, -3.230 ± 0.451.Conclusion: After the building to take in the thick skin wound model ,we successfully constructed the SYBR Green Ⅰ quantitative real-time fluorescence detection method which applied to mRNA of test taking site wound tissue VEGF, TGF-β1 , reveals the alginate dressings importangt role in promoting wound healing , and mainly related to regulation of the expression of cytokines.It provides a reliable theoretical basis to address the reduction of scarring wound healing clinical thorny issues.
