Nature continues to provide a wealth of new leads for potential therapeutic agents or pharmacological probes (1). The role of natural products in traditional medicine is well appreciated and has led to many important drugs that either retain the entire natural product structure or that incorporate (usually straightforward) chemical modifications. Today, biologically useful natural products are most commonly discovered through target-based or phenotypic screening, even in the absence of detailed information about the purpose of the natural product in its original biological setting. The success of these efforts has led some researchers to suggest that scaffolds related to natural product substructures may lead to useful chemical libraries due to the presumed preoptimization of these structures by natural selection (2–4). This argument loses force as compounds become less similar to the natural products that inspired them, but the use of even highly truncated natural product structures in screening can bear fruit if care is taken to retain features consistent with macromolecular binding in the process (5–9). Moreover, it is reasonable to expect that one’s chances for success in identifying structures having activities unrelated to the (possibly unknown) original impetus for evolutionary structure optimization increase if (i) the natural product has multiple bioactivities and (ii) the structure is sufficiently malleable for analogue construction. |

| 自然界不斷為潛在的治療藥物或藥理探針提供新的財(cái)富(1)。傳統(tǒng)醫(yī)藥中的天然成分十分重要,它使得許多重要的藥物要么保留完整的天然的產(chǎn)品結(jié)構(gòu),要么保留合并的(通常是直接的)化學(xué)修飾。今天,即使缺乏天然產(chǎn)品在原初的生物過程中的作用的詳細(xì)信息,人們還是通過基于目標(biāo)或表型的篩選發(fā)現(xiàn)很多生物學(xué)上有用的天然產(chǎn)品。這些努力的成功,使一些研究人員認(rèn)為,根據(jù)這些結(jié)構(gòu)存在自然選擇的預(yù)優(yōu)化的假設(shè),可以將于天然產(chǎn)物結(jié)構(gòu)相關(guān)的基團(tuán)可以組成有用的化學(xué)庫(2–4)。如果化合物變得與其原來的前體不是很一致,這種說法就不是很正確了,但是如果小心的保留與大分子結(jié)合過程相關(guān)的特征結(jié)構(gòu),即使運(yùn)用高度截短的天然產(chǎn)物結(jié)構(gòu)進(jìn)行篩選也能成功(5–9)。此外,如果(i)天然產(chǎn)物有多種生物學(xué)活性,(ii)其結(jié)構(gòu)對于類似物的構(gòu)建具有足夠的延展性,則期望在定義結(jié)構(gòu)方面獲得成功與增加進(jìn)化結(jié)構(gòu)選擇的最初動力(可能是未知的)無關(guān),這也是合理的。 |
