Structural characterizations of P-1 and P-2 (including molecular weight, monosaccharides composition, sulphate and uronic acid contents, conformational structure, ultraviolet spectrum, and infrared spectrum) were investigated, and their anti-proliferation activities against human gastric cancer BGC-823 cells and lung cancer A549 cells were researched in vitro. Results suggested that both P-1 and P-2 could significantly suppress the proliferation of BGC-823 cells in a concentration-dependent manner. The highest inhibitory rates were 78% for P-1 and 95% for P-2 at the concentration of 200 μg/ml, respectively. This high anticancer activity was inferred to be owing to the structural characterizations of polysaccharide, as well as the recognition of biological system for the conformational structure of polysaccharides. |

| 超聲輔助萃取法從草藥A中分離多糖。進一步用DEAE纖維素-52色譜柱與Sephadex G-100分子排阻色譜柱純化獲得P-1與P-2。對P-1與P-2進行了分子鑒定,包括分子量,單糖組成,硫酸與糖醛酸含量,構(gòu)象,紫外譜以及紅外譜。進行了體外抗增生活性實驗,細胞株是人腸道癌BGC-823細胞與肺癌A549細胞。結(jié)果顯示,依據(jù)濃度高低,P-1,P-2能夠顯著抑制BGC-823細胞的增生。P-1,P-2濃度分別達到200 μg/ml時,抑制效率達到最大,78%。如此之高的抗癌活性可能與多糖化學(xué)結(jié)構(gòu)以及生物系統(tǒng)對多糖構(gòu)象的識別有關(guān)。 |

It can be seen from the research that both P-1 and P-2 can significantly inhibit the proliferation of BGC-823 cells in a certain concentration. The highest inhibitory rates were 78% for P-1 and 95% for P-2 at the concentration of 200 μg/ml. This higher anticancer activity may be attribute to the structural characterization of polysaccharides, and the recognition of biological system for the conformational structure of polysaccharides as well . |
