針對(duì)赤霉病導(dǎo)致小麥嚴(yán)重減產(chǎn),而化學(xué)防治和抗病新品種均無(wú)法抑制赤霉病的危害問(wèn)題,以本實(shí)驗(yàn)室發(fā)現(xiàn)的放線菌JK-1聚酮化合物能高效抑制赤霉病菌為契機(jī),采用簡(jiǎn)并引物制備探針、構(gòu)建基因組BAC文庫(kù)獲取PKS基因全長(zhǎng),并進(jìn)行PKS基因的大腸桿菌表達(dá)及農(nóng)桿菌介導(dǎo)的小麥轉(zhuǎn)化表達(dá),研究PKS基因在小麥中成功轉(zhuǎn)化并表達(dá)的技術(shù)難題,探討PKS基因在小麥的抗病作用分子機(jī)理,闡明PKS基因在小麥赤霉病防控方面的重大作用。項(xiàng)目預(yù)期將建立PKS基因轉(zhuǎn)化小麥并成功表達(dá)的技術(shù)體系,揭示PKS基因抗赤霉病的分子機(jī)理。項(xiàng)目的研究結(jié)果對(duì)于獲得比傳統(tǒng)育種及轉(zhuǎn)基因抗病小麥品種更穩(wěn)定而持久的抗赤霉病小麥品種具有重要意義。項(xiàng)目的實(shí)施可以為PKS基因在植物上成功表達(dá)并表現(xiàn)出很好的抗病性提供科學(xué)依據(jù)。 [ Last edited by heihei3910 on 2012-3-5 at 22:11 ] |

Project results for compared with the traditional breeding genetically modified disease-resistant wheat varieties and more stable and enduring resistance varieties of wheat genotypes has important significance. The implementation of the project for the PKS genes in plants express and show successful on very good resistance to provide the scientific basis. |

By taking advantage of the actinomycete JK-1 polyketide found in our laboratory which has proved to be highly effective in inhibiting Fusarium graminearum, the present study, through preparation of probes using degenerate primer and obtaining of full-length PKS genes by constructing a genomic BAC library, completed the PKS gene expression in Escherichia coli and Agrobacterium-mediated transient expression of wheat, solved the technical difficulties in the transformation and expression of PKS genes in wheat, addressed the molecular mechanism of PKS gene’s disease-resistant function, and clarified the significant role PKS genes played in preventing and controlling wheat scab. The project is expected to establish a set of techniques for the transformation and expression of PKS genes in wheat, revealing the molecular mechanism of the PKS gene’s resistance to wheat scab. The research findings of the project shall shed light on obtaining wheat varieties with more stable and lasting resistance to wheat scab, compared with traditional breeding and transgenic disease-resistant wheat varieties. Implementation of the project scientific could provide evidence for the successful expression of PKS genes in plants and their strong resistance to diseases. |
