| 脂氧酶(LOX)為活性中心含鐵的金屬酶。本文將以大豆LOX為模板,研究外源金屬離子對(duì)LOX活性中心的影響及其對(duì)酶活的調(diào)控,驗(yàn)證和完善我們提出的外源金屬離子與LOX活性中心離子存在離子交換的假說(shuō),研究LOX催化過(guò)程中的電子傳遞/離子交換機(jī)制。利用Irving-Williams序列實(shí)現(xiàn)一些外源過(guò)度金屬離子與LOX中鐵的交換,研究其它金屬離子對(duì)LOX的活性或酶促反應(yīng)的影響;采用自制新型指示劑結(jié)合光譜技術(shù)在一定范圍內(nèi)定性定量地研究LOX活性的重構(gòu)對(duì)其結(jié)構(gòu)、活性及催化動(dòng)力學(xué)的影響;實(shí)現(xiàn)對(duì)LOX的金屬修飾并調(diào)控其催化動(dòng)力學(xué)。本文是金屬酶化工的研究和應(yīng)用中亟需解決的共性問(wèn)題,是對(duì)酶化工、天然酶修飾及金屬離子生物效應(yīng)等的應(yīng)用基礎(chǔ)研究;對(duì)了解原生態(tài)金屬酶活性中心金屬離子的作用機(jī)制、內(nèi)外源金屬離子間的相互作用及其活性調(diào)控具有普遍理論意義,也可為研究其它酶促反應(yīng)中金屬離子的作用機(jī)制提供方法上的參考。 |

This is metal enzyme chemical of research and application in the needed solution of common problem, is on enzyme chemical, and natural enzyme modified and the metal ion bio-effect, of application foundation research; for understanding original ecological metal enzyme activity center metal ion of role mechanism, and inside and outside source metal ion between of mutual role and activity regulation has general theory significance, also can for research other enzyme promoting reaction in the metal ion of role mechanism provides method Shang of reference. |
