| 脂氧酶(LOX)為活性中心含鐵的金屬酶。本文將以大豆LOX為模板,研究外源金屬離子對LOX活性中心的影響及其對酶活的調(diào)控,驗證和完善我們提出的外源金屬離子與LOX活性中心離子存在離子交換的假說,研究LOX催化過程中的電子傳遞/離子交換機制。利用Irving-Williams序列實現(xiàn)一些外源過度金屬離子與LOX中鐵的交換,研究其它金屬離子對LOX的活性或酶促反應(yīng)的影響;采用自制新型指示劑結(jié)合光譜技術(shù)在一定范圍內(nèi)定性定量地研究LOX活性的重構(gòu)對其結(jié)構(gòu)、活性及催化動力學(xué)的影響;實現(xiàn)對LOX的金屬修飾并調(diào)控其催化動力學(xué)。本文是金屬酶化工的研究和應(yīng)用中亟需解決的共性問題,是對酶化工、天然酶修飾及金屬離子生物效應(yīng)等的應(yīng)用基礎(chǔ)研究;對了解原生態(tài)金屬酶活性中心金屬離子的作用機制、內(nèi)外源金屬離子間的相互作用及其活性調(diào)控具有普遍理論意義,也可為研究其它酶促反應(yīng)中金屬離子的作用機制提供方法上的參考。 |

LOX is a metalloenzyme with Fe in its active site.
