| 用不同濃度的正己烷或二甲苯處理菌株OS1 和菌株UD2, 發(fā)酵后測(cè)定蛋白酶產(chǎn)量。結(jié)果如圖所示, 不同濃度有機(jī)溶劑脅迫處理均可以顯著提高兩株菌胞外蛋白酶產(chǎn)量�？偟膩�(lái)講，隨著有機(jī)溶劑濃度的增加，蛋白酶分泌量增加，達(dá)到最大值后，增加有機(jī)溶劑濃度，蛋白酶分泌量不發(fā)生顯著變化。10%的正己烷和1%的二甲苯處理OS1以及12.5%的正己烷和5%的二甲苯處理UD2，蛋白酶分泌量達(dá)到最高。有趣的是有機(jī)溶劑脅迫處理不僅可以提高野生型菌株的蛋白酶產(chǎn)量, 也可以提高誘變育種獲得的突變菌株的蛋白酶產(chǎn)量。部分微生物通過(guò)增加胞蛋白酶分泌量提高其有機(jī)溶劑耐受性, 因此,有機(jī)溶劑脅迫會(huì)提高蛋白酶產(chǎn)量。研究者發(fā)現(xiàn)在發(fā)酵培養(yǎng)基中添加有機(jī)溶劑可以提高蛋白酶產(chǎn)量, 但有機(jī)溶劑可能會(huì)造成環(huán)境污染, 并且去除發(fā)酵液中的有機(jī)溶劑相對(duì)困難。用有機(jī)溶劑脅迫處理后發(fā)酵產(chǎn)蛋白酶, 不用在培養(yǎng)基中添加有機(jī)溶劑, 降低成本, 減少工藝, 綠色環(huán)保。 |

| In different concentrations are hexane or deal with OS1 and xylene strains UD2 strains after fermentation determination protease production. In general, with the increasing of the concentration of organic solvent, protease secreted increased to maximum capacity, increase the concentration of organic solvent, protease secreted quantity does not have a significant change. 10% of hexane and 1% of xylene is OS1 processing and 12.5% of hexane and 5% of xylene is UD2 processing, protease secreted reached the highest amount. Interesting is organic solvent handle stress can not only improve the wild type strains protease yield, improve mutation breeding of the strains of protease mutations for production. Part of the microbial by increasing the protease secreted quantity and improve its organic solvent tolerance, therefore, organic solvent stress will improve the protease production. The researchers found that in the fermentation add organic solvent can improve the protease yield, but organic solvents may cause environmental pollution, and remove the fermented liquid organic solvent relatively difficult. Use organic solvent stress after processing ferment produce protease, need not in culture add organic solvent, lower the cost and reduce the process, green environmental protection. |

Deal with bacterial strain OS1 and UD2 with different concentration of n-hexane or xylene , measure the protease yield after fermentation. The results are shown in the figure, stress treatments with different concentration of organic solvent can all greatly improve exocellular protease yield of the two bacteria strains. In summary , the protease yield increased along with the concentration of organic solvent increasing ,but after up to the maximum, , protease yield did not change obviously when increased the concentration of organic solvent. When deal with bacterial strain OS1 with 10% of hexane and 1% of xylene and UD2 with 12.5% of hexane and 5% of xylene, protease yield reached the highest. The interesting thing is that handle stress of organic solvent can not only improve the protease yield of wild bacterial strains, but also improve the protease yield of mutated bacterial strains obained by mutagenesis breeding.
