將鎘處理和非鎘處理的紫葉芥蛋白質(zhì)進(jìn)行分離提純,分析差異蛋白,共得到十三種,本論文著重研究丙糖磷酸異構(gòu)酶(tpiA)。運(yùn)用生物信息學(xué)方法,對(duì)紫葉芥tpiA基因及其編碼蛋白質(zhì)序列進(jìn)行了初步分析得到了基因與蛋白質(zhì)研究相關(guān)的一些基本參數(shù)。結(jié)果表明tpiA基因序列由1268個(gè)堿基組成,編碼長(zhǎng)度為312aa的蛋白質(zhì)。tpiA蛋白分子量大小為35637.5Da,等電點(diǎn)為8.14.。疏水性分析結(jié)果表明tpiA蛋白不具典型的疏水區(qū)域,為親水性蛋白。tpiA具有多種二級(jí)結(jié)構(gòu)形式,其中α-螺旋占33 .29%,無規(guī)卷曲占39.44%,是典型的(α/β)8結(jié)構(gòu)。在SWISS-MODEL服務(wù)器中三維建模后,再在Viewer Lite 4.2軟件中進(jìn)行序列編輯,得到tpiA蛋白的三級(jí)結(jié)構(gòu)。本文還對(duì)tpiA基因進(jìn)行同源基因進(jìn)化分析,結(jié)果顯示紫葉芥的tpiA基因與蕪菁(Brassica rapa)、山崳菜(Eutrema salsugineum)、亞麻薺(Camelina sativa)、薺菜(Capsella rubella)的相關(guān)基因均有較高的同源性。 關(guān)鍵詞:紫葉芥;tpiA基因;生物信息學(xué)分析 |
