內(nèi)源H2S含量測(cè)定方法: 1)試劑 a, 磷酸緩沖液(PH 6.8)50 mM(含0.1M EDTA 和 0.2 M 抗壞血酸 ) b, PBS PH 7.4 100 mM c, L- Cys 10 mM d, 磷酸吡哆醛 2 mM e, 二甲基對(duì)本二胺 5mM(溶入3.5 mM H2SO4) f, 硫酸鐵銨 50 mM (溶入100 mM H2SO4) g, Na2S 溶液(配成已知濃度梯度) 2)繪制標(biāo)準(zhǔn)曲線 稱取Na2S 9H2O 0.2402g 加新沸冷卻的dd water定容至100ml. 取a 將其稀釋為0.5、1、2、4、8、16 μM 測(cè)已知濃度Na2S 670 nm吸光值,繪制標(biāo)準(zhǔn)曲線 3)測(cè)樣品中H2S含量 取0.5 g 樣品在5 ml a中冰浴研磨 將勻漿與b,c,d 混合,室溫放置,離心取上清 將裝有醋酸鋅的吸收井置于裝有上清的小測(cè)試管中,反應(yīng)30 min 加0.3 ml e 加0.3 ml f |

Measurement of H2S content H2S quantification was performed as described by Nashef et al. (1977). The chemical treatments were the same as the measurements of Na+ and K+ contents. Following the treatments, the root of seedlings were ground into fine powder with a mortar and pestle under liquid nitrogen and 0.3 g of frozen tissue were homogenized in 1 ml 100 mM potassium phosphate buffer (pH 7) containing 10 mM ethylenediaminetetraacetic acid (EDTA). The homogenate was centrifuged at 15,000×g for 20 min at 4 °C, and 100 μl of the supernatant was used for the quantification of H2S, in an assay mixture also containing 1,880 μl extraction buffer and 20 μl 20 mM 5,5′- dithiobis (2-nitrobenzoic acid), to a total volume of 2 ml. The assay mixture was incubated at room temperature for 2 min and the absorbance was read at 412 nm.
