NCBI登錄號(hào)申請(qǐng)相關(guān)問題

分離的菌株想在NCBI上申請(qǐng)序列號(hào)，用sequin提交相關(guān)資料后，對(duì)方給我發(fā)回這么一封信： Dear Dr. Huang: We are currently processing your GenBank records JF772465-JF772548 and have come across a point that requires your attention. BLAST similarity search analysis indicates that the following sequence may be misassembled: JF772520 This sequence contains a mixture of plus and minus strand sequence which is biologically incorrect. We have appended an example BLAST result below for your information. Please provide a revised FASTA file with the corrected sequence. In addition, BLAST similarity search analysis indicates that many of your sequences contain little or no similarity to ribosomal RNA sequences in the database at the 5' and 3' ends. Ribosomal RNAs are generally highly conserved and thus, we would expect to see similarity to other rRNA sequences over the entire length. 這是信的內(nèi)容。以前也申請(qǐng)過，但是沒遇到過這個(gè)問題，看不太懂。不知道哪位高手能幫忙解答一下嗎？先謝謝了 |

Please provide a revised FASTA file with the corrected sequence. 這個(gè)說你正負(fù)鏈有拼錯(cuò)的地方，根據(jù)他推薦的seq，看看自己到底哪里錯(cuò)了（如果確定沒問題，可以回復(fù)說就是這樣） In addition, BLAST similarity search analysis indicates that many of your sequences contain little or no similarity to ribosomal RNA sequences in the database at the 5' and 3' ends. Ribosomal RNAs are generally highly conserved and thus, we would expect to see similarity to other rRNA sequences over the entire length. 你的3,5端的序列是測出來的嗎？還是引物加上去的？因?yàn)檫@部分的相當(dāng)保守，而admin認(rèn)為你的seq理由非正常的序列。 你的seq是全長嗎？ 他問到這些問題，之前可能就混過去了，現(xiàn)在不行了，呵呵，大家都很牛的~ ps：對(duì)admin的專業(yè)及敬業(yè)表示欽佩~~~  |
