緊急求助！如何回答reviewer重做實(shí)驗(yàn)的建議

時(shí)逢圣誕期間，以為老外休假，沒(méi)想到今天收到主編回復(fù)，要求大修（所投雜志是Pesticide Biochemistry and Physiology ），和導(dǎo)師討論后意見(jiàn)出現(xiàn)分歧，不知道該怎么辦，向大家求助。 求助問(wèn)題1：有兩個(gè)reviewer回復(fù)意見(jiàn)，都提出語(yǔ)言需要改進(jìn)，老板很堅(jiān)決的說(shuō)不會(huì)花錢找公司改文章，頂多找個(gè)native speaker免費(fèi)修改，這是老板的決定，本人沒(méi)辦法說(shuō)服花錢找公司改。 求助問(wèn)題2：我的研究是尋找一種藥物的作用靶點(diǎn)，其中一個(gè)reviewer提出藥物處理濃度太高，以至于結(jié)果不可信，建議用LD50的40% and 80%的濃度重復(fù)試驗(yàn)。也就是說(shuō)需要所有的實(shí)驗(yàn)重新做，老板也很堅(jiān)決的說(shuō)了，實(shí)驗(yàn)不會(huì)重復(fù)，也不會(huì)補(bǔ)數(shù)據(jù)，因?yàn)樘幚淼乃幬锖吞幚韺?duì)象都沒(méi)有了，不可能再做了。這可怎么辦啊？如果不按照reviewer的建議以低濃度藥物重復(fù)試驗(yàn)，會(huì)不會(huì)直接被拒？？ 以上是主要的問(wèn)題，其他的都是很小的問(wèn)題。先粘貼editor回復(fù)如下： Dear Dr. ###, In regard to your manuscript ###," we now have the reviewer's comments. John Marshall Clark, PhD Pesticide Biochemistry and Physiology Reviewers' comments: Reviewer #1: This manuscript presents interesting results and provides useful methods for molluscicidal mechanism of A on Oncomelania hupensis. However, in my knowledge, the concentrations of A and Nic exposed to snails are too high to obtain the reliable results. The concentrations of 40% and 80% LD50 are much better for the study of molluscicidal mechanism. If possible, the authors should use the proper concentrations to repeat this study. The paper requires a revision of the language and grammar which should be done with the assistance of someone who is more conversant with the language. Reviewer #2: The authors investigated the effect of A on snail mitochondria. The molluscicidal activity of Ais already well established. The present investigations now provide some additional data on the possible mode of action. Generally, the paper is well prepared, the methods applied are appropriate and the conclusions drawn are conclusive.However, there are a number of points which need careful consideration before publication can be recommended: * The paper needs a very comprehensive revision with respect to English grammar and syntax. * Please write ginkgolic acids and niclosamide with a small initial letter throughout the manuscript * The investigations cited in reference [14] were conducted with LLC-MK(2) as well as HaCaT cells. * Page 7, line 1-2: Schistosomiasis joponica should be changed to S. japonica * Page 9: As described in "Statistical Analysis" correlation coefficients were calculated. However, no such results are given. * Page 9: Section 3.1 could entirely be deleted as most of these information are already given in the Materials and Methods section. * Page 10, line 4: LD50 values at 12 h are also shown in Tab. 2 * Page 10, Tab. 2: What statistical test was applied for the calculation of p-values? The legend to Tab. 2 refers to superscripts which do not occur in the Table. * Page 11, line 3: According to Fig. 4 the expression of adenylate kinase does already decrease at 12 h while the mRNA level for cytochrome b seems to drop down only after 24 and 48 h. This disagreement needs to be solved. * Page 11, line 16-18.
