纖維原料是地球上現(xiàn)今最豐富、可再生的廉價的重要資源,對它的應(yīng)用的研究一直是科學(xué)界的重點和熱點。纖維素酶是一種高活性生物催化劑。隨著現(xiàn)代微生物技術(shù)的發(fā)展,纖維素酶在飼料、酒精、紡織和食品等領(lǐng)域顯現(xiàn)出巨大的市場潛力。 實驗室課題組對雙功能纖維素酶CbCel9A1CBM各模塊功能研究后發(fā)現(xiàn)其各截短蛋白間存在漸進(jìn)性和協(xié)同性。為進(jìn)一步具體從結(jié)構(gòu)-功能的關(guān)系了解各截短蛋白,我們希望能夠?qū)bCel 9A-L和L-CBM3C模塊蛋白經(jīng)行晶體結(jié)構(gòu)的解析,從而能從根本上了解其作用機理。首先,我們對目標(biāo)蛋白經(jīng)行了Ni-柱親和分離純化,并通過分子篩層析純化發(fā)現(xiàn)純化效果很好。之后根據(jù)SDS-PAGE凝膠電泳對目標(biāo)蛋白的純度進(jìn)行了考證后用考馬斯亮藍(lán)法對目標(biāo)蛋白的濃度進(jìn)行了測定,并同時濃縮蛋白濃度。之后用懸滴式蒸汽擴散法進(jìn)行大量實驗后,確定了目標(biāo)蛋白的最優(yōu)結(jié)晶條件并進(jìn)行了單晶培養(yǎng)。最后集中送往上海光源中國科學(xué)院上海應(yīng)用物理研究所進(jìn)行X-光同步輻射衍射解析晶體結(jié)構(gòu)。 |

First of all, we have to target proteins in Ni - column affinity purification, and found in a molecular sieve chromatography purification purification effect is very good. Based on sds-page gel electrophoresis after after a textual research on the purity of target protein with coomassie brilliant blue method of target protein concentrations were determined, and at the same time, concentrated protein concentration. After diffusion method using sessile drop type steam after a large number of experiments, the optimal target protein crystallization conditions were determined and the cultivation of single crystal. Finally concentrated to Shanghai light source of Shanghai institute of applied physics, Chinese academy of science for X-ray synchrotron radiation diffraction resolution crystal structure. |
