纖維原料是地球上現(xiàn)今最豐富、可再生的廉價(jià)的重要資源,對(duì)它的應(yīng)用的研究一直是科學(xué)界的重點(diǎn)和熱點(diǎn)。纖維素酶是一種高活性生物催化劑。隨著現(xiàn)代微生物技術(shù)的發(fā)展,纖維素酶在飼料、酒精、紡織和食品等領(lǐng)域顯現(xiàn)出巨大的市場(chǎng)潛力。 實(shí)驗(yàn)室課題組對(duì)雙功能纖維素酶CbCel9A1CBM各模塊功能研究后發(fā)現(xiàn)其各截短蛋白間存在漸進(jìn)性和協(xié)同性。為進(jìn)一步具體從結(jié)構(gòu)-功能的關(guān)系了解各截短蛋白,我們希望能夠?qū)bCel 9A-L和L-CBM3C模塊蛋白經(jīng)行晶體結(jié)構(gòu)的解析,從而能從根本上了解其作用機(jī)理。首先,我們對(duì)目標(biāo)蛋白經(jīng)行了Ni-柱親和分離純化,并通過(guò)分子篩層析純化發(fā)現(xiàn)純化效果很好。之后根據(jù)SDS-PAGE凝膠電泳對(duì)目標(biāo)蛋白的純度進(jìn)行了考證后用考馬斯亮藍(lán)法對(duì)目標(biāo)蛋白的濃度進(jìn)行了測(cè)定,并同時(shí)濃縮蛋白濃度。之后用懸滴式蒸汽擴(kuò)散法進(jìn)行大量實(shí)驗(yàn)后,確定了目標(biāo)蛋白的最優(yōu)結(jié)晶條件并進(jìn)行了單晶培養(yǎng)。最后集中送往上海光源中國(guó)科學(xué)院上海應(yīng)用物理研究所進(jìn)行X-光同步輻射衍射解析晶體結(jié)構(gòu)。 |
